摘 要:目的探讨通光藤提取物抑制人血液肿瘤细胞株增殖及诱导凋亡的作用。方法采用MTT法检测通光藤提取物,包括不同乙醇洗脱组分及C21甾体皂苷单体化合物,作用不同浓度和时间,对人血液肿瘤Raji、NB4和K562细胞增殖的影响;采用流式细胞术检测通光藤C21甾体皂苷单体化合物诱导上述细胞凋亡的作用。结果通光藤70%乙醇洗脱物,在较高浓度(100μg/mL和200μg/mL)下对3种人血液肿瘤细胞株Raji、NB4、K562细胞增殖有显著的抑制作用,优于50%和30%乙醇洗脱物(P<0.05)。4个C21甾体皂苷单体化合物tenacissosides B、C、I和marsdenoside K在体外实验中均表现出对Raji、NB4和K562细胞增殖的抑制作用;其中tenacissoside C的作用最强,和其他3种单体化合物相比差异具有统计学意义(P<0.05),其对Raji、NB4、K562细胞的半数抑制浓度分别为64.1μmol/L、70.4μmol/L和105.8μmol/L。98.4μmol/L tenacissoside C对Raji、NB4和K562细胞分别作用24h和48h,均可促进各肿瘤细胞的早期凋亡和晚期凋亡,与对照组比较凋亡率差异有统计学意义(P<0.05)。结论通光藤提取物,包括不同乙醇洗脱组分及C21甾体皂苷单体化合物,对多种人血液肿瘤细胞株体外有抑制细胞增殖和促进细胞凋亡的作用,其中以C21甾体皂苷单体化合物tenacissoside C的抑制作用最明显,对人Burkitt’s淋巴瘤细胞株Raji增殖的抑制作用最强。Objective To investigate the effects of the extract from Marsdensia tenacissima on proliferation and apoptosis of human hematologic neoplasm cell line cells.Methods Raji,NB4 and K562 cells were treated in vitro with different concentrations of the extract from Marsdensia tenacissima,including different ethanol elution components and C21 steroidal saponin monomer compounds,for different periods.Tumor cell proliferation was measured by MTT colorimetric assay and its apoptosis was determined by the flow cytometry(FCM).Results Firstly,with higher concentrations,100 μg/mL and 200 μg/mL,70% ethanol eluate from Marsdensia tenacissima inhibited the proliferation of Raji,NB4 and K562 cells significantly,in a dose and time dependent manner,compared with 30% and 50% ethanol elution components from Marsdensia tenacissima(P0.05).Secondly,four C21 steroidal saponin monomer compounds,tenacissosides B、C、I and marsdenoside K,also inhibited the proliferation of Raji,NB4 and K562 cells in vitro significantly,in a dose and time dependent manner,compared with that of control group(P0.05).Among them,tenacissoside C showed the strongest inhibition effects on proliferation of these cells under all experimental conditions compared with the other three C21 steroidal saponin monomer compounds(P0.05).Furthermor,the IC_50 of tenacissosides C on proliferation of Raji,NB4 and K562 cells were 64.1 μmol/L,70.4 μmol/L and 105.8 μmol/L,respectively.Finally,after Raji,NB4 and K562 cells were treated with 98.4 μmol/L tenacissoside C for 24 h and 48 h,the early apoptosis rates and late apoptosis rates of these tumor cells increased markedly,compared with the control group(P0.05).Conclusion The extract from Marsdensia tenacissima,including different ethanol elution components and C21 steroidal saponin monomer compounds,may inhibit the proliferation of some human hematologic neoplasm cell line cells and induce these tumor cells apoptosis in vitro,especially tenacissoside C,one of the C21 steroidal saponin monomer compounds
