摘 要:目的探讨中性鞘糖脂在脑胶质瘤细胞生长调控中的作用。方法用改良的Hakomori方法,提取正常脑组织和脑胶质瘤组织中的中性鞘糖脂,并经柱层析法进一步纯化其中的单一成分红细胞糖苷脂(globoside)和CDH,运用扫描电镜观察它们对胶质瘤细胞生长的影响。结果正常脑组织高表达的红细胞糖苷脂可以抑制体外培养人脑胶质瘤细胞株细胞突起的形成,而脑胶质瘤相关CDH对脑胶质瘤细胞生长无明显影响。结论中性鞘糖脂可以抑制胶质瘤细胞突起的形成,其作用与其结构相关。Objective The effects of neutral glycolipids (N-GSLs) on the growth of glioma cells were investigated. Methods The N-GSLs was extracted and purified from normal human brain and glioma tissue with the medified method of Hakomori, globoside and CDH from normal human brain and glioma tissue were further separated from the total N-GSLs by the method of column chromatography. The effects of globoside and CDH on the growth of glioma cells in vitro were observed with scanning electronic microscopy. Results Globoside from normal human brain can inhibit the formation of the protuberance of glioma cells while CDH has no such effect, Conclusions N-GSLs can inhibit the growth of glioma cells, and this ability is closely related to the structure of the N-GSLs.
