摘 要:目的 :明确人表皮生长因子 (human Epidermal Growth Factor,h EGF)对角膜内皮细胞的影响。方法 :采用体外培养的兔角膜内皮细胞 ,观察接种后不同时点 h EGF对其生长状态的影响 ;兔角膜片制作内皮损伤模型 ,离体培养后行 H- E染色和 H3- Td R掺入放射自显影 ,观察 h EGF对其修复的影响。结果 :兔角膜内皮细胞体外培养 h EGF组细胞数高于对照组 ,并使细胞形态产生纺锤形改变 ;角膜内皮损伤 ,离体培养 h EGF组损伤完全修复率高于对照组。结论 :h EGF促进体外培养兔角膜内皮细胞增殖 ;离体培养兔角膜 ,hObjective:To confirm the effects of human epidermal growth factor(hEGF) on corneal endothelial cells.Methods:cultured rabbit corneal endothelial cells were observed and cell counts were determined between experimental group contaning medium hEGF and control group containing medium without hEGF.Freeze thaw injures to rabbit corneas were treated in organ culture.Corneas between experimental group and control group were stained in H-E and used H 3-Thymidine incorporation to determine whether hEGF stimulated repair of rabbit corneal endothelium.Results:after inoculating cells that were exposed to hEGF exhibited a significant increase in growth(P<0 01)compared with control group.Cultured rabbit corneal endothelial cell became spindle shape when they treated in medium with hEGF;The rates of rabbit corneal endothelium totally repaired in experimetal group were highter than those of control group.Conclusion:hEGF promoted the proliferation of the rabbit corneal endotheial cells in vitro and caused a reversible chang in the appearance of the cells;hEGF enhanced wound closure of the rabbit corneal endothelium in organ culture.
