结直肠腺癌中MMP-7的表达与淋巴管生成的关系

摘　　要：目的:通过检测结直肠癌中MMP-7、VEGF-D、VEGFR-3和D2-40的表达,探讨三者的关系以及MMP-7对淋巴管生成的作用。方法:应用免疫组化法检测124例结直肠癌和70例正常黏膜组织中MMP-7、VEGF-D、VEGFR-3和D2-40标记的淋巴管密度(MLVD)的表达,比较两种组织中蛋白表达的差异,分析其与结直肠癌临床病理指标的关系。结果:MMP-7、VEGF-D、VEGFR-3和MLVD的表达在结直肠癌中明显高于黏膜组织(P<0.001),MMP-7的表达与结直肠癌的肿瘤体积、Dukes分期、浸润深度及淋巴结转移有关(P<0.05);VEGF-D、VEGFR-3和MLVD的表达与肿瘤体积、淋巴结转移有关(P<0.05)。结直肠癌中MMP-7和VEGF-D、VEGFR-3、MLVD的表达呈正相关性(均P=0.01)。而且MMP-7和VEGF-D、VEGFR-3、MLVD高表达与患者的不良预后相关(P<0.05)。结论:结直肠癌中MMP-7的高表达可能与结直肠癌中的淋巴管生成有促进作用,联合检测MMP-7、VEGF-D、VEGFR-3 MLVD的表达对判断其预后有一定的指导意义。Objective：To study the expressions of MMP-7,VEGF-D,VEGFR-3 and D2-40 in colorectal carcinomas and relationship between MMP-7 expression and lymphangiogenesis.Methods：Total of 124 cases of colorectal carcinoma and 70 cases of normal colorectal tissues were chosen.The expressions of MMP-7,VEGF-D,VEGFR-3 and D2-40（MLVD） were detected by immunohistochemistry.The clinical characteristic were analyzed.Results：The expressions of MMP-7,VEGF-D,VEGFR-3 and MLVD were higher in the tumour than that in mucosa.The expression of MMP-7 was associated with tumor volume,Dukes stage,invasive depth and lymph node metastasis.The expressions of VEGF-D,VEGFR-3 and MLVD were associated with tumor volume and lymph node metastasis.There was positive correlation between MMP-7,VEGF-D,VEGFR-3 and MLVD in primary tumours.The expressions of MMP-7,VEGF-D,VEGFR-3 and MLVD were correlated with the prognosis of colorectal carcinoma.Conclusion：The overexpressions of MMP-7,VEGF-D and VEGFR-3 might be related to the development and progression of colorectal carcinoma.MMP-7 could be involved in the lymphangiogenesis with VEGF-D and VEGFR-3.MMP-7,VEGF-D,VEGFR-3 and MLVD might be helpful to predict the prognosis in colorectal carcinoma.
