摘  要:以高留胚率水稻辽粳294成熟胚为外植体,对影响高留胚率水稻愈伤组织诱导、分化的接种方式、培养基中激素水平进行研究。结果表明,用解剖刀在胚上纵切1刀后再将种子接在培养基上,种子灭菌彻底,愈伤组织诱导率高且质量较好。诱导培养基中激素6-BA和KT的浓度对愈伤组织的诱导率有极显著的影响,诱导培养基为N6+2 mg/L 2,4-D+0.3 mg/L 6-BA+0.1 mg/L KT时,诱导率较高,可达90.0%;分化培养基为MS+0.8 mg/L NAA+2 mg/L 6-BA时,绿苗分化效果好,分化率为73.0%。Factors affecting the callus induction and differentiation of mature embryo of Liaojing 294 with high plumule ratio were studied. The results showed that the inoculation method of cutting the embryo before inoculating the seed in the medium was an ideal method for a complete sterilization and effective callus induction as the high callus quality could be obtained. The statistical analysis revealed that 6-BA and KT had significant effect on callus induction. The callus induction rate could reach to 90.0% when N6 +2 mg/L 2,4-D+ 0.3 mg/L 6-BA+0.1 mg/L KT was used as callus induction medium. And the cal- lus differentiation rate could be up to 73.0% when using MS+0.8 mg/L NAA+2 mg/L 6-BA as callus differentiation medium.
