脑出血患者抵抗素基因+299G>A位点多态性与血浆抵抗素浓度的相关分析

摘  要:目的探讨浙江地区脑出血患者抵抗素基因+299G>A位点单核苷酸多态性与血浆抵抗素水平的相关性。方法选取344例高血压性基底节出血患者,入院时抽取外周血提取DNA,采用聚合酶反应-限制性片断多态性分析基因型,同时采用ELISA法检测血浆抵抗素浓度,并分析其相关性。结果酶切后可见抵抗素基因+299G>A位点有GG、GA和AA三种基因型。脑出血患者等位基因G和A的频率分别为69.3%和30.7%,基因型GG、GA和AA的频率依次为为49.1%、40.4%和10.5%。脑出血患者各基因型频率符合遗传平衡。GG、GA和AA基因型脑出血患者血浆抵抗素浓度依次为(23.9±7.2)ng/ml、(26.4±7.2)ng/ml和(26.9±9.7)ng/ml。GG基因型患者血浆抵抗素浓度显著低于GA和AA基因型患者(均<0.05),而GA和AA基因型患者血浆抵抗素浓度差异无统计学意义(>0.05)。结论浙江地区脑出血患者抵抗素基因+299G>A位点单核苷酸多态性与血浆抵抗素浓度显著相关,可能参与脑出血炎症反应。Objective To examine the relations of the +299G 〉 A single nucleotide polymorphism ofresistin gene tothe plasma resistin concentrations in intracerebral hemorrhage among Han nationality in Zhejiang province. Methods 344 patients with intracerebral hemorrhage were included in this study. Plasma resistin concentrations were measured,and +299G 〉 A polymorphisms ofresistin gene were genotyped using PCR-restriction fxagment length polymorphism,and then correlation was analyzed. Results The genotypes of the +299G 〉 A polymorphism ofresistin gene includedGG, GA and AA. The allele frequencies of G and A were 69.3% and 30.7%, respectively. The fi'equencies of genotypeGG, GA and AA were 49.1%, 40.4% and 10.5%, respectively. The genotype frequencies of the +299G 〉 A polymorph-ism ofresistin gene were in the Hardy-Weinberg equilibrium. Plasma resistin levels of the patients with the genotype GG,GA and AA were (23.9-J:7.2) ng/ml, (26.4*7.2) ng/ml and (26.9~9.7) ng/ml, respectively. Using analysis of variance, thecommon homozygote (GG) had the lowest resistin plasma concentrations (both P 〈 0.05); the plasma resistin concen-trations between heterozygote (GA) and rare allele homozygote (AA) did not differ significantly (2 〉 0.05). Con-clusions The +299G 〉 A single nucleotide polymorphism ofresistin gene was associated with plasma resistin concen-trations and seemed to be potentially involved in the inflammatory component of intracerebral hemorrhage.
