摘　　要：目的研究APEH基因在肾透明细胞癌中的表达,以探索其意义。方法应用半定量RT-PCR、实时定量PCR、免疫组化(IPH)检测30例配对肾透明细胞癌组织/癌旁正常肾组织标本中APEH基因mRNA和蛋白的表达,并对其中16例配对标本进行Western blot蛋白检测,予以统计分析。结果相较正常对照组,RT-PCR、实时定量PCR及IPH均显示肾细胞癌组织中APEH基因表达下调,其比例分别达到60.0%(18/30)、73.3%(22/30)和80.0%(24/30),另外,Western blot示62.5%(10/16)的癌组织标本APEH蛋白表达下调。4种方法检验P值均<0.05,差异有统计学意义,且针对同一批配对标本各检测方法结果之间一致性显著。结论 APEH可能为一新的肾透明细胞癌分子标志物。Objective To study the expression of APEH in clear cell renal cell carcinoma（ccRCC） and investigate its implication.Methods RT-PCR,real time PCR,immunohistochemistry and related statistical analysis were applied to examine the expression status of APEH in 30 matched ccRCC tumor and adjacent normal kidney tissue samples.Additionally,Western blot was taken to detect the expression of APEH protein in 16 matched tissue.Results When comparing with normal control,there were 60.0%（18/30）,73.3%（22/30） and 80.0%（24/30） of ccRCC specimen were found down-regulated in the detection of APEH expression with the use of RT-PCR,real time PCR,immunohistochemistry respectively.Furtheremore,APEH protein was found down-regulated in 62.5%（10/16） of ccRCC specimen with the application of Western blot.There were obvious difference between the normal control and tumor sample（P0.05）.And all the result of four methods showed a consistency significantly.Conclusions APEH may serve as a potential molecular marker for clear cell renal cell carcinoma.
