摘  要:目的 探讨热疗与放疗联合增强顺铂对子宫颈癌细胞的作用与机制。方法 以体外培养的子宫颈癌Hela 细胞株为研究对象,用组内分组设计方法,以恒温水浴热疗、直线加速器放疗,采用原子吸收光谱法检测Hela 细胞内铂(Pt) 含量及DNA- Pt 含量。结果 10 Gy 以下剂量单独放疗未表现出细胞内Pt 量增加,20 Gy 剂量放疗则其量明显增加,提示放疗损伤细胞膜;37 ℃富氧条件下20 Gy 以下剂量放疗未能增加细胞内DNA- Pt 形成,当辅以43 ℃热疗后,不仅细胞内Pt 含量进一步增加,而且细胞内DNA- Pt 形成也增多,提示热疗能促进放疗损伤的DNA 与Pt 结合。结论 热疗在放疗对顺铂化疗增敏过程中发挥了重要的作用。Objective To study the effect and mechanism of hyperthermia and rediotherapy on uterocervical cancer cells.Methods The experiment was carried out in Hela cells in vitro.The design method was divided into groups in a group to observe their effects.The Hela cells in vitro were treated with hyperthermia(using heat water bath)and radiotherapy(using linear accelerator),the content of intracelluar platinum and DNA-platinum was measured by atomic absorption spectrometry.Results Under the radiotherapy of 10 Gy,the content of intracellular platinum hadn't increased,When adding to 20 Gy,the content was obviously increased.It suggested that cell membrane was damaged by X-rays.However,using single radiotherapy of 20 Gy,the content of DNA-platinum wasn't incerased,but the enhancement of DNA-platinum was found in combination of hyperthermia at 43℃and radiotherapy,besides the increment of intracelluar platinum too.Conclusion It is indicated that,in coordination with the radiotherapy,the hyperthermia plays an important role for treating the Hela cells.
