摘  要:目的:研究抑制Skp2基因表达对NK/T细胞淋巴瘤细胞系HANK1细胞增殖和凋亡的影响,探讨抑制Skp2基因表达治疗NK/T细胞淋巴瘤的潜在应用价值。方法:用RNA干扰(RNA interference,RNAi)方法抑制HANK1细胞Skp2基因表达,实时定量RT-PCR和Western blot检测Skp2基因表达,MTT法检测细胞增殖,流式细胞仪检测细胞周期分布和细胞凋亡。结果:Skp2-shRNA表达载体明显抑制HANK1细胞Skp2基因表达,细胞增殖受抑,细胞周期G1期停滞,凋亡细胞增多。结论:抑制Skp2蛋白功能可能是治疗NK/T细胞淋巴瘤的好方法,值得深入研究。Objectives To study the cell proliferation and apoptosis of NK/T cell lymphoma cell line (HANK1 cell line) after inhibiting Skp2 expression, and investigate the potential role of elimination of Skp2 on the treatment of NK/T cell lymphoma. Methods The expression of Skp2 was inhibited in HANK1 cells by RNA interferance (RNAi) strategy, and the expression of Skp2 was evaluated by Quantitative Real- Time RT-PCR and Western blot, cell proliferation was detected by MTr method, cell cycle distributions and apoptosis were detected by flow cytometry. Results Suppressing Skp2 expression inhibited cell proliferation, caused cell cycle arrested at G1 phase and induced cell apoptosis in HANK1 cells. Conclusions Inhibit Skp2 expression may be a good theraov for oatients with NK/T cell lvmohomas.
