摘 要:观察甘薯提取物对谷氨酸诱导的PC12细胞损伤的保护作用。将大鼠嗜铬细胞瘤细胞(PC12)分为空白对照组、模型组、甘薯提取物0.1、1.0、10.0μg/mL低、中、高剂量组和1.0μmol/L尼莫地平阳性药物对照组,用2.0 mmol/L谷氨酸造成PC12细胞损伤,MTT法测定损伤细胞的存活率,观察其细胞损伤的形态学变化,考察甘薯提取物对谷氨酸所致PC12细胞损伤的保护作用。结果表明甘薯提取物可明显提高谷氨酸诱导的PC12损伤细胞存活率,同时能够明显改善谷氨酸诱导的PC12损伤细胞的细胞形态变化,并呈现剂量相关性,具有一定的神经营养及保护作用。This study investigated the protective action of sweet potato extract to PC12 cell injury induced by glutamie acid. Rat Pheochromocy-'toma cells were divided into control group, model group, low-dose, medium-dose and high-dose sweet potato extract (0.1,1.0 and 10.0 μg/mL) and nimodipine 1.0μmol/L,which were treated by 2.0 mmol/L glu- tamic acid. Survival rate of PC12 ceils were determined with MTr. The results indicated that survival rate was increased and apoptosis rate was declined by sweet potato extract treatment. There was a dose response relationship between surviv- al rate and concentration of sweet potato extract. The results of cytomorphology assay showed that sweet potato extract contributed to improve the PC12 ceil shape change induced by glutamic acid. To sum up, sweet potato extract must have the function of neurotrophic effect and neuroprotection.
