氯沙坦干预心力衰竭大鼠心肌细胞凋亡机制的研究

摘　　要：目的探讨氯沙坦干预心力衰竭大鼠心肌细胞凋亡的机制。方法 24只8周龄SD大鼠随机分为对照组、心力衰竭组及干预组各8只,心力衰竭组采取腹腔注射阿霉素的方式建立模型,干预组在注射阿霉素的同时给予喂服氯沙坦干预,并分别检测各组凋亡指数及促凋亡蛋白Bax和抑凋亡蛋白Bcl-2的表达水平。结果心力衰竭组及干预组细胞凋亡指数均明显高于对照组,但干预组凋亡指数较心力衰竭组明显减低,两者比较有统计学意义(P<0.05);心力衰竭组及干预组Bax、Bcl-2表达均高于对照组,但干预组与心力衰竭组比较其Bax显著降低而Bcl-2显著升高。结论氯沙坦能抑制心肌细胞凋亡,这一作用可能通过上调Bcl-2的表达及下调Bax的表达得以实现。Objective To probe the mechanism of Losartan interfering myocardial apoptosis in heart failure rats.Methods 24 SD male rats（8weeks） were divided into three groups：the control group,the heart-failure group,and the treatment group.Each group had eight rats.We used Adriamycin intraperitoneal injection model,and at the same time the treatment group gavaged to Losartan for six weeks.Finaly,to detect the myocardial apoptosis and the level of Bax protein and Bcl-2 protein 1in all groups.Results The myocardial apoptosis in the heart-failure group and the treatment group were significantly above than the control group,while compared to the heart-failure group,The myocardial apoptosis in the treatment group was significantly decreased;at the same time,the level of Bax protein and Bcl-2 protein in the heart-failure group and the treatment group were significantly more than the control group,but compared to the heart-failure group,the level of Bax protein in the treatment group was less,while the level of Bcl-2 protein was above.Conclusion Losartan can interfer myocardial apoptosis through up-regulating of the level of Bcl-2 protein and down-regulating of the level of Bax protein.
