PGE_2对IL-1α刺激的人牙周膜细胞中MMP-3表达调节作用的研究

摘　　要：目的探讨环氧化酶(COX)-2诱导的前列腺素(PG)-E2对白介素(IL)-1α刺激的人牙周膜细胞(PDL)基质金属蛋白酶(MMP)-3表达的调节作用。方法从牙周组织健康个体获得人PDL细胞,分别在使用和不使用消炎痛(COX抑制剂)、NS-398(选择性COX-2抑制剂)、PGE2的情况下用IL-1α刺激人PDL细胞,通过酶联免疫吸附试验(ELISA)实验分析PGE2水平,通过ELISA和酪蛋白明胶酶谱分析分别评价MMP-3水平和酪蛋白水解活性。结果 IL-1α增加了MMP-3和PGE2的产生。尽管消炎痛和NS-398完全抑制了IL-1α诱导的PGE2的产生,这两种药物却相同程度的加强了IL-1α诱导的人PDL细胞中MMP-3的产生。外源性PGE2以浓度依赖的方式降低了IL-1α诱导的MMP-3的产生。结论COX-2诱导的PGE2抑制人PDL细胞中IL-1α刺激的MMP-3的产生。Objective To investigate whether COX-2-derived PGE2 regulates matrix metallopro- teinase （MMP）-3 production in human periodontal ligament （PDL） eells stimulated with inter- leukin （IL）-lα Methods Human PDL cells obtained from pefiodontally healthy subjects were stimulated with vehicle or IL-loL in the presence or absence of indomethacin （a COX-l/ COX-2 inhibitor）, NS-398 （a specific COX-2 inhibitor）, and PGE2. PGE2 levels were as- sayed by enzyme-linked immunosorbent assay （ELISA）. MMP-3 Revels and caseinolytic activi- ties were evaluated by ELISA and casein zymography, respectively. Results IL-loL enhanced both MMP-3 and PGE2 production. Indomethaein and NS-398 enhaneed IL-1α-induced MMP-3 production in PDL cells, to the same extent, although both the agents completely inhibited IL- ia-induced PGE2 production. Exogenous PGE2 reduced IL-1a-induced MMP-3 production in a dose-dependent manner. Conclusion COX-2-dependent PGE2 downregulates IL-1α-elicited MMP-3 production.
