摘 要:目的:探讨乳腺癌患者白细胞来源微泡(leukocyte-derived microparticles,LMPs)的特性及其临床意义。方法:用流式细胞仪检测44例乳腺癌患者和10例健康志愿者外周血的LMPs以及其中CD44+LMPs、CD24+LMPs和CD44+CD24-LMPs的表达情况。进一步用流式细胞仪检测乳腺癌患者和志愿者外周血单个核细胞(monocytes,MNCs)中CD44+、CD24+和CD44+CD24-细胞的比例。最后分析LMPs的临床相关性以及MNCs与LMPs之间的相关性。结果:乳腺癌组的LMPs、CD44+LMPs和CD44+CD24-LMPs比例显著高于健康对照组(P值分别为0.021、0.011和0.006)。LMPs比例在不同大小的肿瘤组织间差异有统计学意义(P=0.018)。CD44+LMPs比例在不同水平的Her-2表达组间差异也有统计学意义(P=0.020)。MNCs中CD44+、CD24+和CD44+CD24-细胞比例在健康对照组和乳腺癌组之间差异无统计学意义。LMPs与MNCs间无显著相关性。结论:乳腺癌患者的LMPs比例与肿瘤大小有关,而CD44+LMPs比例与Her-2水平密切相关,该指标可用来评估乳腺癌的复发转移及预后情况。Objective:To explore the characteristics of leukocyte-derived microparticles(LMPs) in breast cancer patients and their clinical significance.Methods:The expressions of LMPs,CD44+ LMPs,CD24+ LMPs and CD44+ CD24-LMPs in peripheral blood of 44 breast cancer patients and 10 healthy volunteers were detected by using flow cytometry.The percentages of CD44+,CD24+ and CD44+ CD24-cells in monocytes(MNCs) of breast cancer pateints and healthy volunteers were further detected.The clinical relevance of LMPs,and the correlation between MNCs and LMPs were analyzed.Results:The percentages of LMPs,CD44+ LMPs and CD44+ CD24-LMPs in breast cancer patients were statistically higher than those in the healthy volunteers(P=0.021,P=0.011,and P=0.006,respectively).There was a statistical difference in the percentage of LMPs among the different tumor volunmes(P=0.018).Statistical difference was also obseved in the percentage of CD44+ LMPs among different Her-2 levels(P=0.020).The percentages of CD44+,CD24+ and CD44+ CD24-cells in MNCs in breast cancer patients were not statistically different from those in the healthy volunteers.There was no statistical correlation between MNCs and LMPs.Conclusion:The percentage of LMPs in breast cancer patients can be used to evaluate the tumor volume.The percentage of CD44+ LMPs was closely correlated with Her-2 level,and it can be used to evaluate the recurrence,metastasis and prognosis of breast cancer.
