摘  要:目的:探讨三元复合因子Net对人胰腺癌裸鼠移植瘤生长的抑制作用。方法:以聚合酶链反应(PCR)扩增Net基因全长片段,应用基因重组技术构建Ad5/F35-Net重组腺病毒载体。建立24只人胰腺癌裸鼠移植瘤模型,并随机分为PBS组、绿色荧光蛋白(GFP)组和Net组,3组分别注射0.1 mL的PBS、Ad5/F35-GFP(1×108PFU)和Ad5/F35-Net(1×108PFU),干预3周后,测定各组的移植瘤体积和质量,并应用免疫组化法检测各组移植瘤增殖细胞核抗原(PCNA)表达及移植瘤组织原位细胞凋亡情况。结果:本研究成功构建了表达Net的重组腺病毒载体。与PBS组及GFP组相比,Net组裸鼠移植瘤体积和质量明显减小(P均<0.05),而其PCNA表达显著降低(P<0.01),原位细胞凋亡明显增加(P<0.01)。结论:Net可明显抑制人胰腺癌裸鼠移植瘤的生长,抑制移植瘤细胞的增殖,促进移植瘤细胞原位凋亡。Objective To study the inhibitory effect of the ternary complex factor Net on the growth of human pancreatic cancer transplanted tumor in nude mice.Methods The whole-genome of Net gene was amplified by polymerase chain reaction(PCR),and Ad5/F35-Net recombinant adenovirus vector was constructed by gene recombination.Human pancreatic cancer transplanted tumor model was established in 24 nude mice,then these mice were randomly divided into PBS group,green fluorescent protein(GFP) group and Net group and injected with PBS 0.1 mL,Ad5/F35-GFP 0.1 mL(1×108PFU)and Ad5/F35-Net 0.1 mL(1×108PFU),respectively.Three weeks later,volume and mass of transplanted tumor were measured and expression of proliferating cell nuclear antigen(PCNA) and apoptosis in situ were determined by immunohistochemistry.Results Recombinant adenovirus vector carrying Net was successfully constructed.Compared with PBS group and GFP group,tumor mass,volume,and expression of PCNA in Net group decreased significantly(P〈0.05),and level of apoptosis in situ increased significantly(P〈0.01).Conclusions Net gene could inhibit obviously the growth of human pancreatic cancer transplanted tumor in nude mice,and inhibition of cell proliferation and enhancement of apoptosis might be the mechanism.
