多药耐药相关蛋白-1在难治性颞叶癫痫患者脑组织中的表达及丙磺舒的干预作用

摘  要:目的观察难治性颞叶癫痫患者脑组织中多药耐药相关蛋白1(MRP1)及应用MRP1拮抗剂丙磺舒干预后的表达,探讨MRP1与难治性颞叶癫痫多药耐药的关系。方法应用免疫组化检测难治性颞叶癫痫患者脑组织实验组和对照组MRP1的表达情况,同时应用免疫蛋白印记(westernblot)方法检测实验组、丙磺舒干预组和对照组MRP1的表达。结果免疫组化结果显示MRP1在难治性颞叶癫痫患者脑组织中表达增强,与对照组比较差异有显著性(P〈0.05)。Westernblot结果显示丙磺舒干预组MRP1蛋白水平较实验纽明显较少,差异有显著性(P〈0.05)。结论脑内高表达的MRP1参与难治性颞叶癫痫的耐药机制,丙磺舒可以降低脑组织内MRP1的表达。Objective To investigate the expression of multidrug resistance-associated protein 1 (MRP1) in the brains of patients with intractable temporal lobe epilepsy (ITLE) and the MRP1 expression after application of MRP1 antagonist probenecid, to explore the relationship between MRP1 and drug -resistant mechanism of ITLE. Methods Temporal-lobe specimens from 40 patients with ITLE and brain specimens from 10 patients of similar age and same gender were collected according to the paired design principle as control group. The MRP1 expression in these tissues was detected by immunohistochemistry. The MRP1 expression after probenecid application was detected by Westernblot. Results Compared with the control group,MRP1 expression in brain specimens of patients with ITLE showed a significant increase (P 〈 0.05). MRP1 expression was significantly decreased after inhibition of probenecid (P〈0.05). Conclusions Over expression of MRP1 in the brains of patients with ITLE might contribute to drug-resistant mechanism of medically intractable epilepsy. Probenecid can reduce the expression of MRP1 in brain tissue.
