摘 要:目的对玉郎伞提取工艺进行改进,利用正交法进一步优化提取玉郎伞多糖提取工艺。方法采用正交法优化玉郎伞的提取工艺;以苯酚-硫酸显色法于490 nm波长处测定葡萄糖含量吸光度,计算换算因子和玉郎伞多糖含量。结果葡萄糖的浓度在15~65μg/ml范围内其吸光度呈良好的线性关系(r=0.999 1);平均加样回收率为98.25%,RSD=1.72%(n=6);平均换算因子f=1.82,RSD=2.16%(n=6),玉郎伞多糖的平均提取率为3.78%,RSD=0.173 8%(n=9)。多糖的最佳提取条件为浸泡3 h、加10倍量水、提取3次每次3 h。结论优化提取方法后,玉郎伞多糖的提取率提高98%。苯酚-硫酸法可作为玉郎伞多糖定量分析。Objective To optimize the extraction process of polysaccharides from Yulangsan by orthogonal optimization method. Methods Optimum extraction process for polysaccharides was investigated by orthogonal design.Phenol-sulfuric acid method was used,and the absorbance was measured at the wavelength of 490nm. Results Phenol-sulfuric acid method showed good precision and linearity,the average recovery ratio was 98.25%,and RSD=1.72%(n=6).The optimal conditions were as follows:three hours of water-soaking time,adding ten time amount of water,three hours of extration time and extracting three times. Conclusion Under the optimun conditions,the yield of polysaccharides from Yulangsan can be enhanced by 98%.Phenol-sulfuric acid method can be used for determination of polysaccharides from Yulangsan.
