甘珀酸对癫痫大鼠脑组织缝隙连接蛋白43表达的干预及其意义

摘  要:目的探讨癫痫大鼠脑组织缝隙连接蛋白43(connexin43,Cx43)表达及缝隙连接阻断剂甘珀酸(carbenoxolone,CBX)对其表达的影响。方法健康SD大鼠64只,随机分为正常对照组、致痫组、致痫+生理盐水组(致痫+Ns组)和致痫+CBX干预组(致痫+CBX组)。采用免疫组化染色方法和实时荧光定量逆转录一聚合酶链反应方法分别测定氯化锂一匹罗卡品癫痫模型大鼠脑组织Cx43免疫阳性细胞数及Cx43mRNA表达水平,并观察腹腔注射CBX对Cx43表达的影响。结果与正常对照组比较,致痫大鼠脑组织Cx43免疫阳性细胞数及其mRNA表达差异倍数在致痫1h开始明显增多,并持续至7d(P〈0.05)。大鼠注射CBX后Cx43免疫阳性细胞数及其mRNA表达差异倍数均明显低于同一时间点致痫组和致痫+NS组(P〈0.05),海马区Cx43免疫阳性细胞数及其mRNA表达差异倍数在注射CBX7d后降至正常水平,而皮层区Cx43免疫阳性细胞数及其mRNA表达差异倍数在注射CBX3d后降至正常水平。结论星形胶质细胞Cx43参与了癫痫发作和发展过程。CBX减少致痫大鼠脑组织Cx43表达,具有较明确的抗癫痫作用。To explore connexin (Cx) 43 expression in brain tissues of epileptic rats and the effect of gap junctional blocker carbenoxolone ( CBX ) on Cx43 expression. Methods Cx43 immunoreactive cells and Cx43mRNA expression in brain tissues of lithium chloride piloearpine induced epilepsy model of rats were determined with immunohistochemistry and real time fluorescent quantitative reverse transcriptase polymerase chain reaction (RTPCR) respectively, as well as the effects of intraperitoneal injection of CBX on Cx43 expression were observed. Results Compared with the normal control group, the amount of Cx43 immunoreactive cells and the level of Cx43mRNA expression in brain tissues of epilepsy rat group increased significantly at 1 h after the seizures and lasted for 7 d (P 〈 0.05 ). The amount of Cx43 immunoreactive cells and the level of Cx43mRNA expression in epilepsy + CBX group were significantly lower than those of epilepsy group and epilepsy + saline group at the same time point respectively ( P 〈 0.05 ) , but those in hippocampus decreased to normal level of control group at 7 d after CBX injection, while its expression in cortex recovered to normal level at 3 d after CBX injection. Conclusion Astrocytes Cx43 has participated in the seizure and development of epilepsy. CBX may reduce Cx43 expression in hippocampus and cortex of epileptic rats and show its anti - epilepsy effect.
