摘 要:目的探讨双泛素(FAT10)在原发性骨肉瘤发生发展中的可能作用机制,为以FAT10指标对原发性骨肉瘤的诊断和以FAT10为靶点进行原发性骨肉瘤生物学治疗提供理论依据。方法收集22例原发性骨肉瘤及其骨肉瘤旁骨组织和12例同期接受手术的良性病变骨组织的新鲜标本,采用RT-PCR检测组织中FAT10 mRNA的表达。结果 FAT10 mRNA在原发性骨肉瘤组织、原发性骨肉瘤旁组织和良性病变骨组织中的阳性表达率分别为81.82%(18/22)、13.64%(3/22)和8.33%(1/12)。原发性骨肉瘤旁组织FAT10 mRNA阳性表达率与良性骨组织组织相比,差异无统计学意义(>0.05);原发性骨肉瘤中FAT10 mRNA阳性表达率明显高于原发性骨肉瘤旁组织和良性病变骨组织,差异均有统计学意义(均<0.05)。结论 FAT10mRNA在原发性骨肉瘤中的表达明显上调,其可能成为一个诊断原发性骨肉瘤的检测指标,并为开发骨肉瘤化疗提供新的靶点。Objective To investigate the role of FAT10 in the occurrence and development of primary osteosarcoma,and to provide a new target for biological therapy of primary osteosarcoma.Methods FT-PCR was used to detect the expression of FAT10 mRNA in 22 primary osteosarcoma fresh tissues,22 para-osteosarcoma fresh tissues and 12 benign bone tissues.ResultsPositive rates of FAT10 mRNA in primary osteosarcoma tissues,para-osteosarcoma tissues and benign bone control tissues were 81.82%(18/22),13.64%(3/22)and 8.33%(1/12)respectively.No significant difference was found between para-osteosarcoma bone tissues and benign bone tissues in the positive rates of FAT10 mRNA(P0.05);significant difference was found between primary osteosarcoma tissue and benign bone tissues in the positive rates of FAT10 mRNA(P0.05);significant difference was found between primary osteosarcoma tissue and para-osteosarcoma bone tissues in positive rates of FAT10 mRNA(P0.05).Conclusions Expression of FAT10 mRNA is highly up-regulated in primary osteosarcoma.FAT10 can be used as an index to diagnosis primary osteosarcoma tissues.It may be a new target for biological therapy of primary osteosarcoma.
