摘 要:目的探讨不同剂量直链烷基苯磺酸钠(linear alkylbenzenesulfonate,LAS)致小鼠肝脏损伤作用。方法将40只健康成年ICR种小鼠按单纯随机抽样方法分为正常对照组和LAS低(150 mg/kg)、中(300 mg/kg)、高剂量组(600 mg/kg),采用灌胃方法,持续60 d,每10天称量1次,观察行为,60 d后称量处死,计算肝系数,做病理学检查;检测肝组织中超氧化物歧化酶(super oxidase dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、丙二醛(malondialdehyde,MDA)、乳酸脱氢酶(lactate dehydrogenase,LDH)及一氧化氮合酶(nitric oxide synthase,NOS)几种指标的水平。结果实验过程中,各组小鼠出现不同程度的体质量增长缓慢甚至减轻(P<0.05);低、中、高剂量组肝脏系数分别为(6.25±0.38)%、(6.94±0.52)%和(7.42±0.44)%,均明显高于对照组[(5.32±0.58)%,P<0.05];病理学检查发现有不同程度的损伤,高剂量组肝细胞出现水肿、点状坏死,肝窦充血现象;与对照组相比,各剂量组肝组织MDA含量明显增高,SOD及GSH-Px活性均明显降低(P<0.05,P<0.01);同时,LDH与NOS活性有所增强(P<0.05,P<0.01);结论LAS能抑制小鼠体质量的增长,对肝组织造成一定损伤,影响肝组织抗氧化功能。Objective To investigate the oxidative damage of mouse liver induced by linear alkylbenzenesulfonate(LAS).Methods Forty healthy ICR mice were divided into 4 groups using the method of simple random sampling,that is,control,low-dosage group of LAS(150 mg/kg),middle-dosage group of LAS(300 mg/kg) and high-dosage group of LAS(600 mg/kg) respectively,with every group including 5 male and 5 female mice.The test mice were administered by gavage for 60 d.The mice were weighed every 10 days and their behavior were observed.After 60 days,the mice were killed and liver index were calculated and pathological changes were examined.At the same time,the activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),lactate dehydrogenase(LDH) and nitric oxide synthas(NOS) and the content of malondialdehyde(MDA) were detected.Results LAS treatment at different doses for 60 d resulted a reduce in body weight gain,even in the change of decrease in body weight(compared with the control group,P0.05).Liver index of mice of the different dosage groups was increased [(6.25±0.38)%,(6.94±0.52)% and(7.42±0.44)% compared with the control group(5.32±0.58)%,P0.05].Pathological examination indicated that the damages of liver were found in different degrees.Hydropsia,spotty necrosis and sinusoidal congestion was found in hepatic cells of high-dosage group.The content of MDA and the activity of LDH and NOS in hepatic tissue of LAS groups were increased remarkably,while the activity of SOD and GSH-Px was decreased obviously(compared with the control group P0.05,0.01).Conclusion LAS inhibits the body weight gain,injures hepatic tissue and influences oxidative ability in mice.
