摘  要:目的通过研究大麻素WIN55,212-2对体外培养牛眼小梁细胞(BTMC)环腺苷酸(cAMP)表达的影响,探讨其增加房水排出而降低眼压的机制。方法对BTMC进行体外原代培养及传代培养,应用免疫组化的方法鉴定细胞。然后用不同浓度的WIN55,212-2作用于BTMC,放射免疫法检测细胞上清液中cAMP的水平。结果 WIN55,212-2浓度在0.1-20μmol/L范围内,培养液中cAMP水平随浓度增加而显著性升高,若WIN55,212-2浓度继续增高(40-60μmol/L),cAMP水平不再升高。结论 WIN55,212-2可能通过提高BTMC内cAMP水平而起到降低眼压的作用。To investigate the effect of cannaboid WIN55,212-2 on the expression of cyclic adenosine monophosphate(cAMP) in bovine trabecular meshwork cells(BTMC) in vitro, then discuss its mechanism of reducing intraocular pressure by increasing aqueous humor eduction. Methods BTMC were primary cultured and subcultured in vitro, immunoba'stochemical method were used to identity BTMC. The levels of cAMP in supernatants were measured by radioimmunoassay when BTMC were stimulated by different concertrations of WIN55,212-2. Retlflts When BTMC were stimulated by WIN55,212-2 of concentrations within 0.1-201μmol/L, the levels of cAMP in supernatants significantly increased, but when the concentration further increased(40-60μmol/L), they were no longer rised. Conclusion WIN55,212-2 may play the role of reducing intraocular pressure by increasing the levels of cAMP in BTMC.
