链霉菌S-254菌株产生的凝血酶抑制剂发酵、提取和抑制活力研究

摘　　要：链霉菌S-254菌株产生的凝血酶抑制剂是一种生理活性物质,在发酵过程中抑制剂的产量与发酵培养基的成分有关。在Ⅱ号培养基中,28℃发酵82小时便达到高峰期。发酵液中抑制剂的提取是采用大孔树脂吸附,DFAE纤维素-52、CM纤维素-32离子交换层析和固相化凝血酶亲和层析。经纯化所得到的抑制剂能延长凝血酶时间,对凝血酶呈直接抑制作用。Thrombin inhibitor is a microbial active product, It is produced by streptomyces strain S-254. The output of thrombin inhibitor is related to the composi-ion of medium in the process of fermentation.The production of thrombin inhibitor reached maximum level after fermentation of tie microorganism at 28℃in the No. II medium for 82 hours. The inhibitor in the culture filtrate was purified by absorption on macroporous resin, followed by ion exchenge chromatography on DEAE cellulose -52, CM-cellulose-32 and affinity chromatography on immobilized thrombin.The purified thrombin inhibitor can apparently prolong the thrombin activily and shows that the inhibitor affects thrombin directly.
