摘  要:目的探索冬凌草甲素对人雄激素非依赖性前列腺癌细胞株——PC-3细胞的诱导凋亡作用及其分子机制。方法用不同浓度的冬凌草甲素干预PC-3细胞,MTT试验分析观察其对PC-3细胞活力的影响;流式细胞仪检测细胞周期变化;实时荧光定量PCR方法检测PC-3细胞CyclinD2、CyclinE、P2,蛋白表达的变化。结果(1)冬凌草甲素增加G0/G1期PC.3细胞百分率,降低S期PC-3细胞百分率;(2)冬凌草甲素以浓度依赖性方式抑制PC-3细胞的CyclinD2、CyclinE蛋白表达,而P^27,蛋白表达上调;结论冬凌草甲素通过影响细胞周期调节蛋白、阻断细胞周期G1/S期“稽查点”:抑制CyclinD2、CyclinE,上调P^27等途径抑制PC-3细胞增殖及诱导PC-3细胞凋亡。Objective To investigate the effect of Oridonin on apoptosis of human prostate cancer cell lines (PC-3 cells) and their molecular mechanism. Methods The PC-3 cells were intervened by Oridonin in different concentration. The vitality of the PC-3 cells was detected by MTT assay. The change of cell cycle was analyzed by the flow cytometry; and the changes of expressions of CyclinD2, CyclinE, p27 in PC-3 cells were detected by the real-time fluorescent quantitative determination. Results (1) Oridonin increased the percentage of the G0/G1 phase and decreased the S phase of PC-3 cells; (2) Oridonin down regulated the expression of CyclinD2 and CyclinE, and up regulated the expression of p27 in a concentration-dependent way in PC-3 cells. Conclusion Oridonin can inhibit proliferation of PC-3 cells and induce their apoptosis through regulating the cell cycle protein, blocking the "checkpoint" of the G1/S phase, down-regulating CyclinD2 and CyclinE as well as up-regulating P^27.
