摘 要:目的观察胶质瘤患者肿瘤组织中人类白细胞抗原(HLA)I类抗原及抗原转运相关蛋白(TAP)1、TAP2的表达,探讨其与肿瘤病理级别的关系。方法收集41例经病理确诊的胶质瘤患者肿瘤组织标本,通过免疫组织化学的方法,观察不同病理级别肿瘤标本中HLAI类抗原及抗原加工递呈(APM)分子的表达水平,探讨HLAI类抗原和APM分子表达与胶质瘤患者病理级别之间的关系。结果HLAI类抗原丢失在各级别胶质瘤组织中分别为11%(1/9)、25%(3/12)、30%(3/10)及40%(4/10)。TAPl和TAP2蛋白水平均与胶质瘤分级明显相关(P〈0.05),表达水平随胶质瘤分级增加表达下降。此外,TAP1表达水平与TAP2明显相关(P〈0.05)。结论胶质瘤组织中HLAI抗原和部分APM分子蛋白水平,随胶质瘤级别增加而降低;这可能是导致部分T细胞免疫治疗失败的原因之一。Objective To observe the expression of human leukocyte antigen I ( HLA I) and anti- gen processing machinery (APM) components, and their correlation with pathological grading. Methods Forty-one tumor sections from forty-one glioma patients were examined by using immunohistochemistry. The expression of HLA I antigen and APM components was detected, and their correlation with the pathological grading was analyzed. Results Frequency of HLA I antigen loss in dedicated groups was 11%, 25% , 30% and 40% , respectively. There was a positive correlation between the expression levels of transporter associated with antigen processing (TAP) 1 and TAP2 with glioma pathological grading (P 〈 0.05). Additionally, there was a positive correlation between TAP1 and TAP2 level (P 〈 0.05). Conclusion The level of HLA I antigen and most of APM components expression is decreased with the increase in tumor patho-logical grade, which may explain the phenomenon that glioma cells frequently escape from immunological surveillance or immunotherapy.
