摘  要:目的观察大鼠颅内静脉窦血栓形成(CVST)后脑组织内血管内皮生长因子(VEGF)和VEGF mR-NA的动态表达并其意义。方法采用大鼠上矢状窦结扎并注入脑磷脂白陶土法建立CVST模型,根据CVST后的不同时间分为1d、3d、7d组及假手术对照组。免疫组化法检测VEGF蛋白的表达;RT-PCR法检测VEGF mRNA动态变化。结果 CVST 1d VEGF免疫组化呈显著阳性反应,之后逐渐增强,CVST 7d达高峰;同时CVST后VEGFm-RNA转录水平明显升高。结论颅内静脉窦血栓形成可诱导脑组织内VEGF表达增强,VEGF持续表达可能与促进血管新生和侧支循环形成有关。Objective To observe the expression and change of VEGF and VEGF mRNA in the brain tissue of rats with cerebral venous sinus thrombosis at different time and to study its significance. Methods Methods The superior sagittal sinus of rats was ligated and a kaolin-cephalin suspension was injected into the sinus to establish the CVST model, according to different time after CVST,rats were divided into CVST l d,3d,7d groups and the sham operated control groups. VEGF protein expression was observed by the method of immunohistochemical stain. Dynamic changes of VEGF mRNA were observed using the method of RT-PCR. Results VEGF immunopositive reaction was observed after 1 d of CVST,increased remarkably on 3d, and reached peak on 7d. While VEGF mRNA transcription levels were significantly increased after CVST. Conclusion VEGF expression of the brain tissue enhances after cerebral venous sinus thrombosis,which maybe have a close relationship with the promotion of angiogenesis and the formation of collateral circulation.
