摘 要:目的研究甲基莲心碱对2型糖尿病模型大鼠尿液的代谢产物的影响。方法高脂高糖饲料喂养联合腹腔注射链尿佐菌素造2型糖尿病大鼠模型,糖尿病造模成功后腹腔注射甲基莲心碱20 mg.kg-1.d-1,4周后测大鼠空腹血糖。收集大鼠24 h尿液,样品经处理后进行UPLC/QTOF-MS分析。结果甲基莲心碱处理组大鼠空腹血糖较糖尿病模型组降低(P<0.05)。甲基莲心碱处理糖尿病大鼠后有4种代谢物的含量发生了明显变化,其中二羟延胡索酸(di-hydroxyfumaric acid)下降,前列腺素I2(prostaglandin I2,PGI2)、脂氧素A4(lipoxin A4,LXA4)、脂氧素B4(lipoxinB4,LXB4)升高。结论甲基莲心碱可降低糖尿病大鼠的血糖,改变糖尿病大鼠尿液的代谢产物,其机制可能与其抗氧化应激、抗炎等作用有关。Aim To study whether neferine influences urinary metabolites of type 2 diabetic mellitus rats.Methods The rats were fed with high-sucrose and high-fat chow for 4 weeks,then were injected intraperitoneal(ip) with STZ(30 mg·kg-1).One week later,rats with fasting blood sugar levels higher than 7.8 mmol·L-1 were defined as type 2 diabetes mellitus(T2DM) rats.Neferine treated T2DM rats were injected intraperitoneal(ip) with neferine(20 mg·kg-1·d-1) for 4 weeks.Fasting blood sugar of all rats was measured after 4 weeks.24 h urine samples from rats were collected and prepared.All the separations were performed on UPLC/QTOF-MS system.Results The blood glucose values in neferine treated T2DM rats were decreased in comparison with those in T2DM rats(P0.05).Dihydroxyfumaric acid in urinary metabolite profiles of neferine treated T2DM rats was decreased compared with that in T2DM group.Increase of PGI2,LXA4 and LXB4 was revealed in the urinary metabolite profiles of neferine treated T2DM group than that in T2DM group.Conclusions Neferine can decrease blood glucose and change urinary metabolite profiles of T2DM rats.The mechanism of neferine may be associated with antioxidant stress and anti-inflammatory effect.
