摘  要:目的探讨二烯丙基三硫(DATS)对脂多糖(LPS)诱导急性肺损伤(ALI)小鼠抗氧化功能及细胞间黏附分子-1(ICAM-1)的影响。方法腹腔注射LPS复制小鼠ALI模型,实验动物随机分为生理盐水对照组、ALI组、DATS预防组、DATS治疗组。测定肺系数、肺组织湿/干重量比值(W/D),以及肺组织丙二醛(MDA)含量、髓过氧化物酶(MPO)活性、超氧化物歧化酶(SOD)活性、总抗氧化能力(T-AOC)活性及ICAM-1的表达。结果 ALI组肺系数、肺组织W/D明显升高,肺组织MDA含量增加,MPO活性升高、SOD活性及T-AOC活性降低,ICAM-1表达增加;预防性应用DATS可减轻肺水肿,减少MDA生成,降低MPO活性、增加SOD活性及T-AOC活性,并使ICAM-1表达减少。DATS治疗组与ALI组相比,各指标均无明显变化。结论预防性应用DATS对LPS诱导的ALI小鼠可发挥抗氧化作用,减轻肺损伤程度。Aim To investigate the effect of diallyl trisulfide(DATS) on myeloperoxidase(MPO),malondialdehyde(MDA),superoxide dismutase(SOD),total antioxidant capacity(T-AOC) and intercellular adhesion molecule-1(ICAM-1) in lipopolysaccharide(LPS)-induced acute lung injury(ALI) mice.Methods The mice were randomly divided into saline control group,ALI group,DATS pretreatment group,and DATS treatment group.The lung coefficient and the lung wet/dry weight ratio(W/D) were detected.The activity of MPO,SOD,T-AOC and MDA content in lung tissues were measured.The expression of ICAM-1 in lung tissues was detected by immunochemistry.Results In ALI group after LPS injection,the pulmonary coefficient and W/D were increased.The activity of MPO was increased,whereas the activity of SOD and T-AOC was both decreased.The MDA content and the ICAM-1 expression were also elevated in lung tissues.However,pretreatment with DATS resulted in the decrease of pulmonary coefficient and W/D.Compared with ALI group,the MPO activity,MDA content in DATS pretreatment group were significantly lower,the SOD and T-AOC activity were significantly higher,and the ICAM-1 expression was decreased.But there were no significant differences between DATS treatment group and ALI group.Conclusion Pretreatment with DATS can lighten lung injury through anti-oxidative effect in LPS-induced ALI mice.
