摘  要:目的:探讨血管内皮生长因子(VEGF)和血管性假血友病因子(vWF)在代谢综合征(MS)幼鼠血管壁和血清中的表达及意义。方法:3周龄SD大鼠随机分为普通饮食组(NC组)、高脂组(FC组)和高脂+高盐组(FSC组)。实验4周末测体质量、腹围、血压、内脏脂肪质量、血脂,行口服葡萄糖耐量试验,计算胰岛素抵抗指数。取腹主动脉免疫组化法检测血管壁VEGF和vWF表达,ELISA方法检测血清VEGF和vWF表达。结果:FSC组体质量、腹围、血压、内脏脂肪、血糖、胰岛素水平均较NC组增加,血脂紊乱加重,并出现胰岛素抵抗。FC组仅体质量、内脏脂肪质量高于NC组,其他上述指标差异无统计学意义。FSC组血管壁及血清中VEGF和vWF较NC组和FC组均升高。FC组较NC组血管VEGF及血清中VEGF和vWF均升高。结论:VEGF和vWF参与了血管壁的功能紊乱或损伤的病生理改变。Objective:To investigate the expression of vascular endothelial growth factor (VEGF) and Von Willebrand?factor (vWF) in vascular wall and serum of metabolic syndrome in young rats. Methods:Three-week-old SD rats (50 g body weight) were randomly divided into 3 groups, routine diet group (NC), high fat diet group (FC) and high fat + high salt diet group (FSC). The values of body weight, abdominal circumference, blood pressure, visceral fat weight and plasma lipids were measured in rats 4 weeks after experiments. The oral glucose tolerance test (OGTT)was performed and the insulin resistance index was calculated. The expressions of VEGF and vWF in the abdominal aorta were detected by immunohistochemistry method. The serum levels of VEGF and vWF were detected by ELISA method. Results: The values of body weight, abdominal circumference, blood pressure, visceral fat, plasma glucose and insulin level were significantly higher in FSC group than those of NC group. There were increased dyslipidemia and insulin resistance in FSC group compared with those of NC group. The values of body weight and visceral fat were significantly increased in FC group than those of NC group. The expression levels of VEGF and vWF in abdominal aorta serum were significantly higher in FSC group than those of NC and FC groups. The serum expressions of VEGF and wWF were significantly increased, but not for the vWF expression in abdominal aorta, in FC group than those of NC group. Conclusion: The high expression of VEGF and vWF may be related to the vascular pathological and physiological changes of metabolic syndrome in rats.
