摘  要:目的观察重组人血小板活化因子乙酰水解酶(rPAF-AH)对小鼠脑梗死后基质金属蛋白酶9(MMP-9)表达的影响。方法选择昆明小鼠100只,分层随机分为对照组、模型组、rPAF-AH组、金纳多组和奥扎格雷钠组,每组20只。采用线栓法建立小鼠大脑中动脉缺血再灌注模型,尾静脉注射rPAF-AH预处理后,观测模型小鼠神经行为学和脑梗死灶体积的改变,采用Western blot法比较MMP-9的表达及活性。结果与模型组比较,rPAF-AH组小鼠2、24 h神经行为学评分明显降低(P<0.01),脑梗死灶体积明显减小(P<0.01)。Western blot结果显示,与模型组比较,rPAF-AH组小鼠MMP-9表达明显降低(P<0.01)。结论重组人PAF-AH预处理可减轻小鼠脑梗死损伤,这可能得益于其抑制了MMP-9蛋白的高表达。Objective To observe the effect of matrix metall proteinase 9(MMP-9) on expression of recombinant human platelet activating factor aeetylhydrolase(rPAF-AH)in mice following cerebral infarction. Methods One hundred Kunming mice were randomly divided into control group, model group,rPAF-AH group,ozagrel sodium group, and Ginkgo Biloba leaf extract group(20 in each group). A mouse cerebral middle artery ischemia/reperfusion model was egtablished. Neurological behavior defect and cerebral infarction size in model group were observed after pretreatment with rPAF AH injected through the coceygeal vein. MMP-9 expression and activity levels were measured by Western blot. Results The neurological behavior defect score was significantly lower and the cerebral infaction size was significantly smaller in rPAF-AH group than in other groups(P〈0.01). Western blot showed that the MMP-9 expression and activity levels were significantly lower in rPAF-AH group than in other groups(P〈0.01). Conclusion Pretreatment with rPAF-AH can improve cerebral infarction in mice by inhibiting the over-expression of MMP-9.
