摘 要:目的探讨代谢酶基因C Y P 1A1和G STM 1多态性与广西人群胃癌遗传易感性之间的相关性。方法采用PC R技术检测广西地区121例胃癌患者和138例健康人的C Y P 1A1基因M SP1多态性和G STM 1基因多态性的分布频率,分析其与广西地区胃癌遗传易感性的相关性及交互作用。结果 C Y P 1A1、M SP1 3种基因型(m 1/m 1、m 1/m 2、m 2/m 2)分布频率在两组间比较无统计学意义(χ2=0.901,P=0.342)。胃癌组G STM 1(-)基因型频率(54.5%)显著高于对照组(39.1%)(χ2=6.140,P=0.013)。携带G STM 1(-)基因型的个体患胃癌的风险是携带G STM 1(+)基因型个体的2.13倍(95%C I=1.079~1.831,P=0.013)。与同时携带G STM 1(+)和C Y P 1A1 M SP1 m 1/m 1的个体比较,同时携带G STM 1(-)和C Y P 1A1 M SP1突变m 2基因型的个体患胃癌的O R为2.327(95%C I=1.110~4.280),差异具有统计学意义(P=0.030)。结论单独的C Y P 1A1基因M SP1多态性与广西人群胃癌易感性无明显相关。G STM 1(-)基因型增加广西人群患胃癌的风险。G STM 1(-)基因型与C Y P 1A1 m 2突变基因型联合显著增加广西人群的胃癌易感性。【Objective】To evaluate the relationship between genetic polymorphisms of CYP 1A1 MSP1 and GSTM1 genes and gastric cancer susceptibility in population from Guangxi Province of China.【Methods】A case control study was carried out to compare the distribution frequencies of CYP 1A1 MSP1 and GSTM1 gene polymorphisms between gastric cancer(n =121) and control healthy individuals(n =138) in population from Guangxi Province of China by polymerase chain reaction and to analyze the associations between genetic polymorphisms and susceptibility of gastric cancer.【Results】The frequency of CYP 1A1 MSP1 genotypes(m1/m1,m1/m2,m2/m2) in gastric cancer group and control group was 35.5%,43.0%,21.5% and 41.3%,39.1%,19.6%,respectively.There was no significant difference between the two groups(χ2 =0.901,P =0.342).The frequency of GSTM1 null genotype in gastric cancer group was significantly higher than that in control group(54.5% vs 39.1%,χ2 =6.140,P =0.013).The individuals who carried with GSTM1 null genotype had a 2.13 fold increased risk of gastric cancer(95% CI=1.079~1.831,P =0.013) than those carried with GSTM1(+) genotype after data was stratified by age,gender,smoking status and drinking status.Compared with people who had the combination of GSTM1(+) genotype and CYP 1A1 m1/m1 genotype,The odds ratio of gastric cancer for the combination of GSTM1 null genotype and at least one CYP 1A1 m2 genotype was 2.327(P =0.030).Combination of GSTM1 null genotype and at least one CYP 1A1 m2 genotype significantly increased gastric cancer susceptibility.【Conclusion】There is no obvious relationship between the CYP 1A1 MSP1 polymorphisms and gastric cancer in Chinese Guangxi population.GSTM1 null genotype is closely associated with increased risk for gastric cancer in Chinese Guangxi population.The combination of GSTM1 null genotype and CYP 1A1 m2 genotype can increase gastric cancer susceptibility in Chinese Guangxi population.
