摘 要:目的:探索低剂量电离辐射对LAK细胞治疗动物骨肉瘤的影响。方法:培养的UMR-106细胞接种在月龄大鼠的前胸壁皮下,建立动物骨肉瘤模型。将LAK细胞分别经不同剂量X线辐照后,分别于3、6、9、12、15、18和21d进行细胞计数。采用3H-TdR释放法测定LAK细胞的杀伤活性。winn氏测定法测定LAK细胞对皮下肿瘤细胞生长的影响。结果:大鼠肿瘤发生率达到100%,动物模型建立成功。LAK细胞在第6~9天时,3.25mGy受照组细胞扩增量明显大于对照组。经0.65、3.25mGy辐照及未辐照的LAK细胞输入荷瘤小鼠后,其生存期以3.25mGy组为最长。将骨肉瘤细胞与LAK细胞一并注入小鼠右后肢皮下,30d后发现,3.25mGy辐射的LAK细胞小鼠未见肿瘤生长,而0及0.65mGy辐照的LAK细胞小鼠,可见质量2.0g肿瘤生长,对照组为6.25g。结论:低剂量电离辐射可以使LAK细胞扩增,低剂量电离辐射后LAK细胞的杀伤活性及对骨肉瘤的抗瘤作用均明显增强。OBJECTIVE: To explore the influence of low-dose ionizing radiation for treating animals osteosarcoma with LAK cells.METHODS: The cultivated UMR-106 cell was inoculated under the anterior chest wall skin of month aged mice,and the animal osteosarcoma model was created.Putting LAK cells under the different doses of X ray,and then counting the number of cells after 3,6,9 and 12,15,18,21 days with different doses of X ray.Measuring the lethal activity of LAK cells with 3H-TdR Release method,and measuring the LAK cells influence on subcutaneous tumor cells with WINN Determination method.RESULTS: The tumor incidence rate of elder mice was 100%,and the animal model was successfully created.From the sixth to the nineth day,the LAK cells proliferation of 3.25 mGy irradiation group was significantly clearer than the control group.Among the different tumor-bearing mice injected with 0.65 mGy,3.25 mGy and non-radiated LAK cells,the survival period of 3.25 mGy group was the longest.Thirth days after jnjecting osteosarcoma cells and LAK cells into mice right leg subcutaneous tissue together,it turned out that no tumor proliferation could be seen in 3.25 mGy irradiation group,but 2.0 g tumor appeared in 0 and 0.65 mGy irradiation group with LAK cells,at the same time the proliferation tumor weight of contract group is 6.25 g.CONCLUSION: Low-dose ionizing radiation can stimulate the proliferation of LAK cells,and after low-dose ionizing radiation,the LAK cells lethal activity and its anti-tumor effect to osteosarcoma is significantly enhanced as well.
