摘 要:目的:本研究拟探讨绿色荧光蛋白转染是否可导致心脏L-型钙电流发生变化。方法:在小鼠心脏上注射表达增强型GFP(EGFP)的腺病毒,一周后分离心肌细胞,用膜片钳记录ICa的大小及通道动力学过程。结果:转染EGFP可显著增大细胞膜电容。与对照组相比,EGFP组ICa的电流密度、激/失活动力学参数没有明显变化,但ICa的失活后恢复时间加快。结论:转染EGFP可导致心肌细胞肥大,但对ICa的大小、激/失活动力学过程不产生影响,其加快ICa失活后恢复的作用可能与CaMKII的磷酸化无关。Objective: To speculate that green fluorescent protein(GFP) maybe alter L-type calcium current characteristics in myocardium. Methods: Adenovirus integrated with enhanced green fluorescent protein(EGFP)was injected in mouse hearts.A week later,the mouse hearts were digested and the single cells were used to record ICa by patch clamp technique. Results: In EGFP group,the cell capacitance increased significantly compared with that in control group.ICa density was similar between control and EGFP group,and no change was found in parameters of activation and inactivation.The recovery from inactivation was obviously accelerated in EGFP group. Conclusion: Transfection of EGFP can induce cardiac hypertrophy,but not alter ICa density and activation/inactivation process.EGFP can accelerate ICa recovery from inactivation,but this influence may not be related to CaMKII activity.
