摘 要:目的 采取手术标本 ,在裸鼠皮下连续传代建立人肾母细胞瘤裸鼠移植瘤模型。方法 切取肾母细胞瘤患儿肿瘤组织植入裸小鼠右前肢皮下 ,连续传代。观测肿瘤生长 ,对各代移植瘤进行光镜和电镜观察 ,波形蛋白 (vimentin)及上皮膜抗原 (EMA)免疫组化染色 ,WT1基因检测。结果 10个月中连续在裸鼠皮下传 4代 ,总成瘤率为 43%。移植瘤生长曲线与Gompentz函数符合。移植瘤保留了原肾母细胞瘤的组织学特征。Vimentin在各代肿瘤细胞中均可见表达。而EMA在本肿瘤中均无阳性表现。图像分析结果提示各代移植瘤DNA指数及其细胞时相分布基本一致。WT1基因3段短重复序列长度在各代之间无变化。结论 该模型基本保留了原肿瘤的生物学特性 ,是肾母细胞瘤基础及临床研究的良好模型。Objective To establish Wilms' tumor model in nude mice by hypodermic inoculation of human Wilms' tumor specimens.Methods Fresh human Wilms' tumour specimens were sliced and inoculated hypodermically into the forelimbs of nude mice. The tumor proliferation was monitored. Tumor samples were subjected to light and electronic microscopy examination and were immunostained for Vimentin and EMA. WT1 gene expression was evaluated with PCR.Results During the 10 months period of study, 4 generations of tumor divisions were recorded. The growth curve corresponded to the Gompentz function. The transplanted tumor cells retained the original histological tumor character. Immuno-staining of Vimentin was positive in each generation of tumor cells whereas that of EMA was negative. DNA index and cell cycle distribution were similar in each generation. The 3' end of WT1 gene sequence remained intact in each generation.Conclusion The nude mice Wilms' tumor model retains all the biological characteristics of the tumor and is suitable for Wilms' tumor study.
