错配修复基因hMSH2蛋白在人卵巢恶性肿瘤组织中的表达及其临床意义

摘　　要：目的　检测hMSH2基因在人卵巢恶性肿瘤组织中的表达及其与卵巢恶性肿瘤临床病理特征的关系。方法　应用免疫组织化学链酶菌抗生物素蛋白 过氧化物酶连接法检测 42例卵巢恶性肿瘤组织中错配修复基因hMSH2蛋白的表达。结果　 2 6例卵巢恶性肿瘤组织有hMSH2蛋白表达(6 2 % ) ,多发生于浆液性腺癌和粘液性腺癌 (80 %和 80 % ) ,而在子宫内膜样癌和恶性生殖细胞肿瘤中表达较少 (1/ 6和 1/ 6 ,P <0 .0 5 ) ,其中中～高分化癌hMSH2蛋白的表达率为 83% ,高于低分化癌(46 % ) (P <0 .0 5 )。无淋巴结转移者hMSH2蛋白的表达率 (72 % ) ,高于有淋巴结转移者 (31% ,P<0 .0 5 )。hMSH2蛋白的表达与卵巢恶性肿瘤的临床分期无明显相关性。结论　hMSH2基因参与了卵巢恶性肿瘤的发生、发展及分化过程 ,并与预后有密切的关系。hMSH2蛋白的检测有助于卵巢恶性肿瘤的早期诊断及判断预后。Objectives TostudytheexpressionofhMSH2geneinhumanmalignantovariantumorandits relationshiptoclinicopathologicalcharacteristics .Methods TheexpressionofhMSH2genein 42patientswith malignantovariantumorwasdetectedbyimmunohistochemistrystreptomycinperoxidelmethodusingmonoclonal antibodytohMSH2protein .Results hMSH2expressionwasobservedinthe 2 6of 42malignantovariantumors (6 2 % ) ,morefrequentlypresentinserousandmucinousadenocarcinoma (80 % ,80 % )thaninendometrioid adenocarcinomaandmalignantgerminaltumors (1/ 6 ,1/ 6 ) (P <0 .0 5 ) .Itwassignificantlyhigherinhighgrade thaninlowgrade (P <0 .0 5 ) ,andthereducedexpressionofhMSH2wasalsosignificantlymoreofteninlymph nodesmetastasizedcases.StatisticalanalysisindicatedthathMSH2expressionwasn′trelatedtothestageofthe tumors.Conclusions TheseresultssuggestthattheabnormalexpressionofhMSH2mightbeinvolvedinthe carcinogenesisprocessandprogressionofmalignanttumor,andthatthelossofthehMSH2proteinexpressionmight beausefulpredictorofearlydiagnosisandprognosisofmalignantovariantumor.
