摘  要:目的 :虎奶菌碱溶性多糖的分离纯化及成分分析。方法 :用 0 .5mol·L-1氢氧化钠溶液从虎奶菌核中提取多糖 ,利用气相色谱法 (GLC)测定多糖组成 ,凝胶渗透色谱法 (GPC)测定相对分子质量 ,红外光谱法 (FT IR)测定糖苷键构型 ,高碘酸氧化及Smith降解确定糖基间连接方式。结果 :虎奶菌多糖由葡萄糖组成 ,平均相对分子质量为 4.3× 10 5。经高碘酸氧化 ,每摩尔单糖残基消耗 0 .48mol·L-1高碘酸 ,同时释放 0 .2 4mol·L-1甲酸 ,氧化产物经还原 ,水解后检出葡萄糖和甘油。红外光谱在890cm-1处有吸收 ,而在 840cm-1处无吸收。结论 :虎奶菌碱溶性多糖为 (1→ 3) β D 葡聚糖 ,并含 1,6 连续支链。OBJECTIVE:To isolate the alkali soluble polysaccharide from the scierotia of Pleurotus tuber regium (Fr.) Sing .(or Hunai) and analyze the composition of the polysaccharide. METHODS:To extract the polysaccharide with sodium hydroxide solution.The composition was determined by GLC of the aldononitrile acetates of the monosaccharides resulted from the complete hydrolysis of the polysaccharide.The molecular weight was estimated by GPC.The forms of glycoside linkage were determined from the results of periodate oxidation,Smith degradatin and infrared (FT IR) spectroscopy.RESULTS:The polysaccharide (HNP) was composed of glucosyl residues and the molecular weight was ca.4.3×10 5.After the completion of the periodate oxidationl,peniodate consumed and formic acid liberated were 0.48 mol and 0.24 mol for per mole of glucosyl residues,respectively.In the hydrolyzate of the periodate oxidized product,glucose and glycerol were detected by PC and GLC.An absorption at 890 cm -1 in the FT IR spectra in dicates the β configuration. CONCLUSION:The lkali soluble polysaccharide (HNP) from the sclerotia of Hunai was defined as a (1→3) β D glucan with branched at O 6.
