奈韦拉平对甲状腺未分化癌细胞增殖及钠-碘同向转运体表达的影响

摘　　要：目的探讨非核苷类逆转录酶抑制剂奈韦拉平对甲状腺未分化癌细胞FRO增殖及细胞中Na^＋-I^-同向转运体（NIS）表达的影响。方法以不同浓度的奈韦拉平诱导FRO细胞，观察细胞增殖情况；FRO细胞在用奈韦拉平处理48h后利用实时荧光定量PCR检测细胞中NIS和促甲状腺激素受体（TSHR）基因的表达。结果奈韦拉平诱导后，FRO细胞增殖受到抑制；200和350μmol／L奈韦拉平诱导48h后，FRO细胞中NISmRNA的表达分别是对照组的（1．39±0．04）和（1．85±0．28）倍（均P〈0．01），TSHRmRNA表达分别是对照组的（2．23±1．47）和（2．83±0．78）倍（均P〈0．05）。结论奈韦拉平能够抑制FRO细胞增殖，并且能够诱导细胞中NISmRNA和TSHR mRNA表达增加。Objective To explore the effects of nevirapine on the proliferation and expression of sodium-iodide symporter （NIS） mRNA in FRO cells in vitro. Methods The cells were incubated in different concentrations of nevirapine to evaluate the cell growth rate. And the expressions of NIS mRNA and TSHR mRNA were determined by real-time quantitative reverse polymerase chain reaction. Results Cell proliferation was inhibited after a 96 h nevirapine treatment. After incubating in the presence of 200 and 350 p.mol/L nevirapine, the expression of NIS mRNA was （ 1.39 ± 0. 04 ） and （ 1.85 ± 0. 28 ） times versus the control cells （P 〈 0. 01 ） while the expressions of TSHR mRNA was （2. 23 ± 1.47 ）and（ 2. 83 ± 0. 78 ）times versus the control cells respectively （ both P 〈 0. 05）. Conclusion Nevirapine inhibits cell proliferation in FRO cell line. More importantly, the cells exposed to nevirapine may induce the up-regulations of NIS mRNA and TSHR mRNA.
