卵圆细胞标志分子在肝癌细胞株BEL-7402中的表达及其意义

摘　　要：目的探讨肝癌细胞株BEL-7402的发生与卵圆细胞之间的分子关联及诱导分化前后的变化特征。方法应用免疫细胞化学和酶细胞化学技术检测卵圆细胞的多种标志分子CK7、CK8、CK18、CK19、AFP及GGT在人肝癌细胞株BEL-7402中的表达状态,并观察BEL-7402诱导分化前后各种标志分子的表达变化特征。结果 BEL-7402中CK7、CK8、CK18、AFP及GGT表达阳性,CK19表达阴性,与卵圆细胞向肝细胞分化早期阶段的分子表达谱相似。BEL-7402诱导分化前后各分子标记物及GGT酶活性的表达无明显改变。结论 BEL-7402与卵圆细胞在分子表达谱上有高度相似性,提示肝细胞癌的发生与卵圆细胞之间存在重要关联。在诱导分化过程中,BEL-7402标志分子表达水平的变化要晚于其在细胞周期方面的变化。Objective To detect the molecular association between oval cells and the genesis of cell line BEL-7402 of human hepatocellular carcinoma,as well as the variation characteristics of BEL-7402 cells before and after differentiation-inducing.Methods The expression status of oval cell markers CK7,CK8,CK18,CK19,AFP and GGT in BEL-7402 cell line was detected by means of immunocytochemistry and enzyme cytochemistry techniques,and the change of expression level of the molecules was observed before and after differentiation-inducing.Results The results showed that all the detected markers were positively expressed in BEL-7402 cell line,with the exception of CK19.It was similar to the early molecular spectrum of oval cell differentiating towards hepatocyte.However,no distinct change of expression level of the molecules and the enzyme activity of GGT could be observed in BEL-7402 cell line before and after differentiation-inducing.Conclusion The great similarity of the molecular spectrum between oval cells and BEL-7402 cells suggested that there must be an important relationship between oval cells and the genesis of hepatocellular carcinoma.The change of expression level of the marker molecules in BEL-7402 cells might be later than the changes in cell cycle during the process of differentiation-inducing.
