清开灵体外抗氧化活性与机制研究

摘　　要：目的:本文旨在研究清开灵注射液的抗氧化活性。方法:通过建立XO/XTT氧自由基生成体系和黄嘌呤氧化酶致尿酸生成反应,以及次黄嘌呤/黄嘌呤氧化酶诱导的细胞氧化损伤模型,分别研究不同浓度清开灵的自由基清除能力,对黄嘌呤氧化酶活性的干预作用和对氧化损伤细胞的保护作用。结果:清开灵具有良好的清除氧自由基作用,并可显著抑制黄嘌呤氧化酶活性,对细胞氧化损伤模型显示明显的保护作用,可显著提高细胞生存率。结论:清开灵注射液具有良好的抗氧化活性,该作用与清除自由基和抑制黄嘌呤氧化酶活性有关。Objective: To study the antioxidant activity of Qingkailing injection.Methods: Through the establishment of XO/XTT oxygen free radical production system and the uric acid formation reaction caused by xanthine oxidase and the cellular oxidative damage model induced by xanthine/xanthine oxidase,the free radicals scavenging ability of different concentrations of Qingkailing,the intervention role of the xanthine oxidase activity and the protective effect of oxidative damage to cells were studied respectively.Results: Qingkailing had a scavenging effect of the oxygen free radical,significantly inhibited the activity of xanthine oxidase,obviously protect the cells oxidative injury model and improved the rate of cell survival.Conclusion: Qingkailing injection has good antioxidant activity,which is related to the free radical scavenging and the inhibition of xanthine oxidase activity.
