摘 要:目的环介导等温扩增法(loop-mediated isothermal amplification,LAMP)是一种新型等温核酸扩增方法。由于其具有简便、高效、特异性高和成本低等优点,在甲型H1N1流感和肺结核等流行病检测中得到了广泛应用。在LAMP技术中,关键的起始步骤是设计合适的引物序列。为了让引物设计更加方便与高效,我们开发了LAMP引物设计软件BioSunLAMP。方法采用Delphi程序设计语言开发了界面友好、便于使用的软件系统。结果经甲型H1N1流感、结核分枝杆菌实验验证,BioSunLAMP软件设计的引物达到了预期效果。此外,与同类软件相比,BioSunLAMP还具有如下特点:①集引物设计与引物特异性分析于一体,可以通过本地数据库或远程调用NCBI的相关数据库来检查引物特异性;②支持针对多序列的通用引物与特异引物设计。结论 BioSunLAMP软件的开发,为LAMP技术的普及提供了很好的生物信息学支持。Objective Loop-mediated isothermal amplification(LAMP) is a novel approach to nucleic acid amplification.Due to its user-friendliness,high specificity,efficiency and low cost,LAMP method is widely used for detecting infectious diseases such as H1N1 flu and tuberculosis.The key initial step for LAMP method is to design a set of proper primers.In order to make primer design easier,we developed a software system,BioSunLAMP,for primer selection.Methods Delphi was employed to develop the user-friendly and easy-to-use software.Results BioSunLAMP was successfully applied to design the primers for the H1N1 flu and tuberculosis.Moreover,compared to other similar tools,BioSunLAMP had the following features.First,primer design and specificity checking were integrated into one system.In addition,the local nucleotide database or remote NCBI nucleotide database could be called for primer specificity check.Second,it could be used to design common and specific primers for a set of closely-related multiple sequences.Conclusion BioSunLAMP provides better bioinformatics support for the wide application of LAMP method.
