摘  要:目的研究在下颌骨发育过度导致的骨性Ⅲ类错畸形患者中,胰岛素样生长因子-1(IGF-1)基因的表达,探讨IGF-1基因表达与下颌骨生长的关系。方法选取骨性Ⅲ类错畸形患者27例,骨性Ⅰ类患者(包括个别正常)27例,应用荧光定量PCR技术和酶联免疫吸附剂测定(ELISA)技术研究下颌骨中IGF-1基因的表达变化。结果 RT-PCR检测结果显示,骨性Ⅲ类错畸形组中IGF-1 mRNA的表达量为(5.541 0±2.044 7)μg/μl,骨性Ⅰ类组中IGF-1 mRNA的表达量为(1.282 1±0.273 1)μg/μl,2组比较具有统计学差异(P<0.05)。根据IGF-1基因光密度值与浓度关系曲线,得回归系数0.998。ELISA检测结果显示,骨性Ⅲ类错畸形组中IGF-1蛋白浓度为(84.125 9±29.294 7)ng/L,骨性Ⅰ类组中IGF-1蛋白浓度为(22.406 4±4.931 2)ng/L,2组相比具有显著差异(P<0.05)。结论下颌骨生长量越大,IGF-1基因mRNA和蛋白表达越高。Objective To study the expression of IGF-! in bony class m malocclusion patients and its relationship with growth of mandible. Methods Twenty-four bony class ]lI malocclusion patients and 27 bony class [ malocclusion patients were included in this study. Expression of IGF-1 in mandible tissue was detected by RT-PCR and ELISA, respectively. Results RT-PCR showed that the expression level of IGF-1 mRNA was significantly higher in bony class ]lI malocclusion patients than in bony class [ malocclusion patients (5. 541 0 _+ 2. 044 7 i^g/txl vs 1. 282 1 +0.273 1 txg/txl, P 〈0. 05) . The regression factor was 0.998, which was calcu- lated according to the correlation curve for IGF-1 optical density value and its concentration. ELISA showed that the expression level of IGF-1 protein was significantly higher in bony class malocclusion patients than in bony class [ malocclusion patients (84. 125 9 +_29.294 7 ng/L vs 22.406 4 +4.931 2 ng/L, P 〈0. 05). Conclusion The more the mandible grows, the higher the expression level of IGF-1 is, which indicates that IGF-1 plays an important role in growth of mandible condyloid cartilage.
