重组苦瓜子抗HIV植物蛋白30对BGC-823细胞增殖与凋亡的影响

摘　　要：目的探讨重组苦瓜子抗HIV植物蛋白30(MAP30)对人胃癌BGC-823细胞增殖与凋亡的影响。方法将BGC-823细胞分为对照组(A组)和MAP30 10、20、30、40、50、60μg/ml实验组(分别为B1、B2、B3、B4、B5、B6组)。MTT、集落形成和裸鼠致瘤实验观察细胞增殖的抑制作用,光镜、电镜、荧光染色和流式细胞术检测细胞凋亡。结果与A组相比,MAP30呈剂量依赖性地抑制BGC-823细胞增殖,IC50为51.67μg/ml。B3组光镜下细胞变圆缩小,细胞膜发泡,可见凋亡小体;电镜下细胞表面微绒毛消失,细胞核和细胞质浓缩,核膜褶皱,染色质凝集。荧光染色和流式细胞术检测显示,B3组BGC-823细胞凋亡率较A组增加(P<0.05)。结论 MAP30可抑制BGC-823细胞增殖,并可诱导其凋亡。Objective To investigate the effect of recombinant Momordica anti-HIV protein of 30 kD（MAP30） on the proliferation and apoptosis of human gastric carcinoma BGC-823 cells.MethodsThe BGC-823 cells were divided into groups of A（normal controls）,B1（treated with MAP30 10 μg/ml）,B2（treated with MAP30 20 μg/ml）,B3（treated with MAP30 30 μg/ml）,B4（treated with MAP30 40 μg/ml）,B5（treated with MAP30 50 μg/ml） and B6（treated with MAP30 60 μg/ml）.The inhibitory effect on cell proliferation was determined by MTT assay,colony formation and tumor xenograft model of nude mice.The cell apoptosis was detected by light microscope,electron microscope,fluorescence staining and flow cytometry.Results Compared with group A,MAP30 inhibited the proliferation of BGC-823 cells in a dose-dependent manner and the IC50 was 51.67 μg/ml.In group B3,some cells showed round and smaller in size,membrane blebbing and formation of apoptotic body under light microscope,meanwhile the disappeared microvilli on the surface of the cells,nucleus and cytoplasm shrinkage,nuclear membrane fold and chromatin agglutination were observed under electron microscope.The fluorescence staining and flow cytometry indicated that the apoptosis rate of BGC-823 cells in group B3 was higher than that in group A（P〈0.05）.ConclusionMAP30 can inhibit the proliferation of BGC-823cells and induce the cell apoptosis.
