核小体结合蛋白调控CXCR4抑制前列腺癌PC-3细胞的侵袭作用

摘　　要：目的探讨抑制人核小体结合蛋白1(NSBP1)基因后抑制非激素依赖性前列腺癌细胞系PC-3侵袭能力的作用机制。方法通过慢病毒lentivirus-NSBP1转染非激素依赖性前列腺癌细胞系PC-3后,MTT法观察细胞活性变化,Transwell实验观察细胞侵袭能力的变化,应用Western-blot技术检测NSBP1、CXCR4蛋白的变化情况。结果抑制NSBP1表达水平后非激素依赖性前列腺癌细胞系PC-3细胞活性降低,侵袭能力降低,同时伴有CXCR4蛋白的表达降低。结论通过慢病毒转染抑制NSBP1的表达可以抑制非激素依赖性前列腺癌细胞系PC-3的侵袭能力,NSBP1可能是通过调控CXCR4蛋白的变化影响细胞侵袭能力的。Objective To study the effect and mechanism of nucleosome-binding protein 1（NSBP1）modulating invasion on human androgen-independent prostate cancer cell PC-3.Methods This study employed lentivirus mediated knockdown of NSBP1 to elucidate the oncogenic role of NSBP1 in prostate cancer PC-3 cell,MTT assay and transwell assay were used to observed cell proliferation and invasion,Western-blot was used to test the expression of NSBP1 and CXCR4.Results The transfected lentivirus-NSBP1 PC-3 cells induced decrease of cell proliferation and invasion,and at the same time the expression of CXCR4 protein also decreased.Conclusions Knocking down of NSBP1 induce decrease of cell proliferation and invasion,which may have role in SDF-1/CXCR4 ways.
