了哥王中西瑞香素对照品的制备及鉴定

摘　　要：目的建立从了哥王[Wikstroemia indica(L.)C.A.Mey]茎皮中制备西瑞香素对照品的方法。方法采用萃取、硅胶柱层析和重结晶方法对酶水解后了哥王茎皮的甲醇提取物进行分离纯化,通过MS、IR、1H-NMR和13C-NMR进行结构鉴定,通过TLC法和HPLC法对对照品进行纯度检测。结果从了哥王茎皮中分离纯化出西瑞香素对照品,质量分数>99.0%。结论该方法制备的西瑞香素对照品符合中药化学对照品的相关要求,可作为了哥王药材及其制剂质量控制用的化学对照品。Objective To establish a method for the preparation of daphnoretin′s reference substance from the stem bark of Wikstroemia indica（L.） C.A.Mey.Methods Daphnoretin was isolated and purified from the MeOH extract of the stem bark of Wikstroemia indica by enzyme hydrolysis,solvent extraction,silica gel column chromatography and re-crystallization.Its structure was identified by MS,IR,1H-NMR and 13C-NMR.Its purity was detected by thin layer chromatography（TLC） and high pressure liquid chromatography（HPLC）.Results Daphnoretin was obtained from the stem bark of Wikstroemia indica with a purity of more than 99.0%.Conclusion Daphnoretin prepared by this method can meet the demands of chemical reference substances in Traditional Chinese Medicine,which is useful in the quality control of Wikstroemia indica and its preparations.
