摘 要:目的探讨Bcrp1+宫颈癌Hela细胞的侵袭及转移能力。方法采用免疫荧光标记物流式细胞仪分选HeLa细胞系中Bcrp1+表型细胞,并利用Boden小室侵袭试验检测两组Bcrp1+表型及Bcrp1-表型细胞的侵袭能力。结果 Hela细胞系中存在Bcrp1+细胞,比例为7.1%。Bcrp1+表型组侵袭过人工基底膜的细胞数为99±14.2,侵袭深度为(365.61±52.32)μm;Bcrp1-表型组侵袭过人工基底膜的细胞数为57±13.3,侵袭深度(301.04±53.81)μm。Bcrp1+亚群细胞穿透人工基底膜的细胞数及穿透深度均明显强于Bcrp1-亚群,差异有统计学意义(P<0.05)。结论Bcrp1+表型Hela细胞具备较强的侵袭转移能力,其中可能含有宫颈癌干细胞,Bcrp1可能成为宫颈癌干细胞标记物。Objective To identify the ability of migration and invasion of Bcrpl^+ phenotype Hela cells and in order to make sure if Berpl is the marker of cervical cancer stem cells or not. Methods Methods Flow cytometry was used to isolate the Bcrpl ^+ cells in Hela cell line , tumor cell migration and invasion were measured by Boyden chamber to identify the ability of Bcrpl ^+ phenotype Hela cells. Results Hela cells contained 7.1% Bcrpl ^+ cells. The number of invasive cells under the chamber of Bcrpl ^+ cells was 99 ± 14. 2, the depth of invasion through matrigel was (365.61 ±52. 32) μm. The number of invasive cells under the chamber of Bcrpl - cells was 57 ± 13.3, the depth of invasion through matrigel was (301.04 ± 53.81 )μm. The difference between the two groups was significant. Conclusions Bcrpl ^+ cells have more migration and invasion ability than Bcrpl^- cells. Bcrpl might be the marker of cervical cancer stem cells.
