美洲大蠊提取液对溃疡性结肠炎大鼠结肠IL-4,bFGF表达的影响

摘　　要：目的 通过观察美洲大蠊提取液（ACE）对溃疡性结肠炎（UC）大鼠的疗效及结肠黏膜中白细胞介素4（IL-4）、碱性成纤维细胞生长因子（bFGF）表达的影响,从抗炎及黏膜修复方面阐明ACE治疗UC的作用机制.方法 2,4,6-三硝基苯磺酸（TNBS）灌肠法制备UC模型.将36只健康雄性Wistar大鼠随机分为对照组10只与TNBS组26只.造模第3天随机处死TNBS组大鼠6只,观察造模效果.将TNBS组剩余的20只大鼠随机分为模型组与实验组,每组10只.实验组于第3天起给予ACE治疗,模型组与对照组给予等量生理盐水.造模第13天,对大鼠行疾病活动指数（DAI）评分,处死大鼠,收集标本,行结肠大体形态评分及组织学损伤评分,分别用免疫组织化学法及实时荧光定量PCR技术观察大鼠结肠黏膜中IL-4,bFGF的表达.结果 与对照组相比,实验组与模型组DAI评分、结肠大体形态评分、组织学损伤评分均升高,而实验组比模型组显著改善（P〈0.05）;与对照组相比,实验组与模型组IL-4表达显著降低,而实验组又比模型组显著升高（P〈0.01）.与对照组相比,模型组、实验组bFGF表达显著升高,而实验组又比模型组显著升高（P〈0.01）.结论 ACE对UC大鼠疗效显著,其作用机制之一可能是通过上调结肠黏膜IL-4,bFGF的表达来抗炎及促进黏膜的修复.Objective To evaluate the effect of American cockroach extract （ ACE ） and the levels of interleukin-4 （1L-4） and basle fibroblasl growth frocfor（bFGF） in colonie mueosa and to study the function and mechanism of ACE in rat.,.; with ulcerative colitis. Methods Thirty six Wistar rats were divided ranctmly into control group（ 10） and TNBS group（26）. Ulcerative colitis was induced in Ite TNBS group bv rectal administration of trinitrobenzene sulphonie acid（TNBS）. After three days,six rats in TNBS group were killed randomly. Then the remanenl rats in TNBS group were divided randomly into model group and experimental group. Experimental group was treatmented with ACE. Control group and model group ere treatmented with sodium chloride. After ten clays,all rats were killed. Disease activity index（ DAI ） mucosal lesion area and histological damage scores were assessed,and the expression of IL-4 and bFGF in the colonic mtteosa were detected by real-time fluorescent quantification polyrerase chain reaetin and immunohistoehemislry.. Results Compared with the. model group, the DAI, mucosal lesion area and histological damage scores in experimental group were significantly improve（ P 〈 0. 05） ;Compared with model grcup,the levels of IE-4 and bFGF in experimental group were significantly increased（ P 〈0. 01 ）. Conclusion ACE ean ameliorate colonic inflammation in rats with ulcerative colitis possibly by increasing the expression of IL-4 and bFGF in the colonic mucosa.
