人牙周膜干细胞的多向分化潜能实验

摘　　要：目的:体外培养分离人牙周膜干细胞(hPDLSC),做成骨诱导和成脂诱导实验。方法:取新鲜拔除正畸牙的牙周膜为材料制成组织块,加入培养液以组织块法分离培养,免疫磁珠法筛选人牙周膜干细胞,加入成骨诱导液诱导牙周膜干细胞成骨,加入成脂诱导液诱导牙周膜干细胞向脂肪细胞转化。结果:成功筛选出人牙周膜干细胞,成骨及成脂诱导成功。结论:免疫磁珠法是筛选人牙周膜干细胞的有效方法,牙周膜干细胞可以向成骨细胞和脂肪细胞分化。Objective： To induce the periodontal ligament stem cell in vitro into osteoblast and adipocyte. Method： Pe- riodontal ligament tissue was been obtained from extracted orthodontal dental, then cultivated with nutrient solution, hPDLSC was selected by immunomagnetic beads.hPDLSC was induced into osteoblast in the osteogenic nutrient solution,was induced into adipocyte in the adipogenic nutrient solution. Result： hPDLSC was selected successfully with immunomagnetic beads method.hPDLSC was induced successfully into osteoblast and adipocyte. Conclusion： Immunomagnetic beads method was an effective way to isolate and purify the hPDLSC and hPDLSC was able to transfer into osteoblast andadipocyte.
