摘 要:目的:研究血管内皮细胞对卵巢癌淋巴结高转移细胞的运动迁移和侵袭能力的影响。方法:建立卵巢癌淋巴结高转移细胞(SKOV3-PM4)、人脐血管内皮细胞(human umbilical venous endothelial cells,HUVEC)条件培养基的交互培养体系,通过MTT法测定生长曲线,HE染色观察细胞超微结构的改变,Transwell穿膜运动实验、划痕实验、明胶酶谱分析等实验方法研究交互培养后各组细胞的侵袭转移能力的变化。结果:与同期单独培养的SKOV3-PM4、HUVEC相比,交互培养后的SK-OV3-PM4伪足增多,生长速度明显快于SKOV3-PM4,交互培养后HUVEC呈不规则形态,出现空泡化超微结构,生长速度与HUVEC相似,细胞的运动和侵袭能力增强,基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)在单独的SKOV3-PM4中低表达,在HUVEC中不表达,在共培养的SKOV3-PM4-HUVEC中高表达。结论:HUVEC上清液培养后的SKOV3-PM4的生物学行为和运动侵袭能力发生改变,推测可能与血管内皮分泌的VEGF、MMP-2等因子有关。Objective: To establish the condition medium of culture cell system with SKOV3-PM4 and HUVEC and to study their invasiveness and movement capability.Methods: To collect the culture medium of SKOV3-PM4 cells as SKOV3-PM4 condition medium and the culture medium of HUEVC as HUEVC condition medium.The growth curve of the cells was determined by MTT assay.The ultrastructure and morphological changes of cells in the condition medium cultivation were detected by Hematoxylin and eosin staining.The change of each cells invasiveness and migration capability were detected by healing assay,transwell chamber model and gelatin zymography.Results: As compared to the single culture SKOV3-PM4,the cells which cultured in HUVEC condition medium showed the increase of pseudopodia and grows more quickly,the healing assay and transwell chamber model showed that invasiveness and migration capability enhancement.Compared to the single culture HUVEC,the cells which cultured in SKOV3-PM4 condition medium showed the morphological significantly changed and vacuolization in the cytoplasm and grew more slowly,the healing assay and transwell chamber model showed that invasiveness and migration capability enhancement.Gelatin zymography assay showed that MMP-2 low-expressed in SKOV3-PM4 and didn't expressed in HUVEC in RPMI1640 medium and high-expressed in SKOV3-PM4-HUVEC in the co-culture system.Conclusion: The biological characteristics and invasiveness capability of SKOV3-PM4 show significantly changes in HUVEC condition medium of culture,it may related to vascular endothelial secretion of VEGF,MMP-2 and other factors.
