固本抑瘤Ⅱ号及其拆方含药血清诱导MCF7细胞自噬的研究

摘　　要：目的:研究固本抑瘤Ⅱ号(GYⅡ)及其拆方大鼠含药血清对人乳腺癌细胞MCF7自噬的影响。方法:制备GYⅡ及其拆方大鼠含药血清,培养MCF7细胞,四甲基偶氮唑盐(MTT)法检测细胞生长抑制,单丹磺酰尸胺(MDC)及Hoechst 33342双染色荧光显微镜、微管相关蛋白1轻链3(MAP-LC3)蛋白Ⅱ(MAP-LC3-Ⅱ)染色激光共聚焦及透射电镜观察自噬,MAP-LC3-Ⅱ/DNA双参数流式细胞术同步分析细胞自噬与细胞周期。结果:与空白组比较,全方组、益气组含药血清处理MCF7细胞表现出细胞抑制效应,活血组、鸡血藤组含药血清处理MCF7细胞均表现出明显的自噬。结论:GYⅡ全方和益气组具有抑制MCF7细胞增殖的效应,而活血组和鸡血藤组未见抑制作用,可能与该两组含药血清诱导MCF7细胞自噬有关。Objective： To observe autophagy induced by Guben Yiliu Ⅱ（GY Ⅱ） and its disassembled recipes medicated serums on MCF7 cells.Methods： GY Ⅱ and its disassembled recipes medicated serums were prepared,MCF7 cells were cultured.The tumor cells inhibition was measured by methyl thiazolyl tetrazolium（MTT）assay.Autophagy was observed by the fluorescent dye monodansylcadaverine（MDC） and Hoechst 33342.Microtubule associated protein 1 light chain 3 Ⅱ（MAP-LC3-Ⅱ） expression was observed by flow cytometric and confocal laser microscope.Electron microscopy has been used to demonstrate the enrichment of lysosomal,autophagic vacuoles and autophagosomes.Results： Compared with the control group,GY Ⅱ group and tonifying Qi group＇s treatment on MCF7 cells showed inhibitory effects,promoting Blood circulation group and Millettia dielsiana group＇s treatment on MCF7 cells showed a significant autophagy.Conclusion： GYⅡ group and tonifying Qi group inhibit the proliferation of MCF7,while promoting blood circulation group and Millettia dielsiana group showed limited cells inhibition,which may be related to autophagy on MCF7 induced by the medicated serum.
