摘  要:目的 通过观察槲皮素对宫颈癌 HeLa 细胞中midkine(MK)、caspase-3 基因和蛋白表达水平,探讨槲皮素体外诱导 HeLa 细胞凋亡的机制.方法 宫颈癌细胞株 HeLa 细胞经培养、传代后分为实验组(分别加入浓度为10、20、40、80及160 μmol/L的槲皮素)和空白对照组.采用逆转录-多聚酶链反应(RT-PCR)和蛋白质印迹(Western blot)方法检测细胞内的MK和caspase-3基因mRNA和蛋白表达水平.结果 随槲皮素浓度的增高HeLa细胞中MK mRNA和蛋白表达依次下降,其中80 μmol/L、160 μmol/L组下降较明显,而caspase-3的mRNA和蛋白表达随药物浓度增高表达增强(P〈0.01).结论 槲皮素可能通过下调MK、上调caspase-3的表达水平而发挥抗宫颈癌作用.Objective To explore the influence of quercetin against cerical cancer possible mechanism of the apoptosis in vitro by observing the effects of quereetin (Que) on Midkine (MK) and caspase -3 genes and protein expression level of the human uterine cervix cancer HeLa cells. Methods HeLa cells cultured in vitro were divided into two groups: the experimental group and the control group. In experimental group, the cells were respectively treated with quercetin at different concentrations,10 μmol/L, 20 μmol/L, 40 μmol/L, 80 μmol/L and 160 μmol/L, while the cells in the control group weren't treated with quercetin. The expression of MK and easpase -3 mRNA and protein in HeLa cells was measured by reverse transcription(RT) - PCR and Western blot. Results Along with the increase of Que con- centration, MK mRNA and protein level down -regulated, in 80μmol/L and 160μmol/L group was significantly lower, but caspase - 3 mRNA and protein level obviously up - regulated ( P 〈 0.01 ). Conclusion Que can induce the apoptosis of tumors through down - regulating MK expression and activating caspase - 3 expression.
