摘  要:目的采用皮下注射蛙皮素诱导急性胰腺炎大鼠(AP)模型,观察中介素(IMD)在AP中的变化,并通过腹腔注射外源性IMD8-47,观察其对胰腺的保护作用。方法①选取雄性SD大鼠24只,体质量250~300g,随机分成3组:正常对照组(NS)、急性胰腺炎6h(AP-6h)组和12h(AP-12h)组,皮下注射蛙皮素(100μg/kg)制备AP模型,采用蛋白印迹技术检测各组胰腺组织中IMD蛋白含量变化;②雄性SD大鼠32只,体质量250~300g,随机分成4组:NS组、AP组、肾上腺髓质素(ADM)治疗组、IMD8-47治疗组,同上述方法制备AP模型,2治疗组分别于造模后2h腹腔注射ADM、IMD8-47(7nmol/kg)。通过烘干法检测各组胰腺组织水肿程度,比色法检测血清中淀粉酶(AMY)、诱导型一氧化氮合酶(iNOS)、内皮型一氧化氮合酶(eNOS)、一氧化氮(NO)值的变化。结果①蛋白印迹检测结果:AP-6h组和AP-12h组IMD比对照组明显减少(P〈0.01);②胰腺组织水肿程度:AP组〉ADM组〉IMD8-47组〉对照组,且AP组与治疗组和对照组间差异有统计学意义(P〈0.05),2治疗组之间差异无统计学意义(P〉0.05);血清中iNOS、NO、AMY:AP组〉ADM组〉IMD8-47组〉对照组,且各组间差异有统计学意义(P〈0.05);Objective To determine the variation in intermedin (IMD) in rats with acute pancreatitis (AP) by establishing the caerulein-induced AP model via subcutaneous injection of caerulein, and to investigate the protective effect of exogenous IMD8-47 intraperitoneal injection on the pancreas. Methods A total of 24 male SD rats weighed 250~300 g were randomly assigned into normal control (NS group), 6-hour acute pancreatitis (AP-6 h group) and 12-hour acute pancreatitis group (AP-12 h group), and were subject to subcutaneous caerulein injection (100 μg/kg) for preparation of AP model. The variation in IMD level in pancreas tissues, as assessed via Western blotting, was com-pared among groups. We then randomly divided 32 male SD rats weighed 250~300 g into normal control (NS group), acute pancreatitis (AP group), adrenomedullin treatment (ADM group) and IMD8-47 treatment group (IMD8-47 group), re-spectively. The approach for preparing the model of AP was described previously. Both treatment groups underwent ADM and IMD8-47 (7 nmol/kg) injection intraperitoneally 2 hours after establishment of AP model. Pancreatic edema was evaluated by using the drying method, and the variation in serum amylase (AMY), inducible nitric oxide synthase (iNOS), endothelial nitric oxide synthase (eNOS) and nitric oxide was assessed via colorimetric assay. Results Both AP-6 h and AP-12 h group, as revealed by Western blotting, exhibited markedly lower (P0.01) IMD level than nor-mal controls. For pancreatic edema, the degree was ordered as AP groupADM groupIMD8-47 groupcontrol group, and in addition, there was significant difference between AP group and treatment group as well as between AP group and control group (both P0.05), but not between both treatment groups (P0.05). For serum iNOS, NO and AMY, the levels followed the order of AP groupADM groupIMD8-47 groupcontrol group (P0.05).
