摘 要:目的观察青蒿琥酯对人结肠癌HCT-8细胞诱导血管新生的影响,并探讨其作用机制。方法采用大鼠主动脉环体外培养实验和鸡胚绒毛尿囊膜体内血管生长实验观察青蒿琥酯对人结肠癌HCT-8细胞诱导血管新生的影响。采用Western blot法检测青蒿琥酯作用前后人结肠癌HCT-8细胞的血管内皮生长因子(VEGF)、血管生成素-2(Ang-2)蛋白。结果青蒿琥酯可明显减少HCT-8细胞诱导的血管生成。HCT-8细胞高表达VEGF、Ang-2蛋白,青蒿琥酯呈剂量依赖方式下调HCT-8细胞VEGF、Ang-2蛋白的表达(P均<0.05)。结论青蒿琥酯具有抗人结肠癌HCT-8细胞诱导血管新生的效应,与青蒿琥酯抑制HCT-8细胞的VEGF、Ang-2蛋白表达有关。Objective To investigate the inhibitory effect mechanism of artesunate on angiogenesis induced by human colon cancer HCT-8 cells, and study the mechanism. Methods The aortic sprouting in fibrin gel experiment and chick embryo choriailantoic membrane (CAM) experiment was used for the research of artesunate on angiogenesis induced by hu- man colon cancer HCT-8 cells. After artesunate treatment, the VEGF and Ang-2 in human colon cancer I-ICT-8 cells were studied by Western blot. Results Artesunate significantly reduced the human colon cancer HCT-8 cells-induced angiogen- esis (P 〈 0. 05). The VEGF and Ang-2 was over expression in the human colon cancer HCT-8 cells, and artesunate could down-regulate the levels of VEGF and Ang-2 in human colon cancer HCT-8 cells in a dose-dependent pattern ( all P 〈 0. 05). Conclusion Artesunate can significantly inhibit the angiogenesis induced by human colon cancer HCT-8 cells, which may be related to the decreasion in the expression level of VEGF and Ang-2 after artesunate treatment.
